Camptothecin effectively treats obesity in mice through GDF15 induction

Jun Feng Lu et al.

Summary: In this study, we found that the small molecule Camptothecin (CPT) can induce the production of GDF15, which has anti-obesity effects through activation of the GDF15-specific receptor GFRAL pathway. This discovery provides a new direction for the development of obesity treatments.

The GDF15-GFRAL pathway is dispensable for the effects of metformin on energy balance

Anders B. Klein et al.

Summary: Metformin has been widely used, but the mechanism of its weight loss effect is not fully understood. This study confirms that metformin can increase the levels of GDF15 in the body, but the role of the GDF15-GFRAL pathway in mediating the weight loss effect of metformin remains controversial. The results suggest that the effects of metformin on energy balance may not solely depend on the GDF15-GFRAL pathway.

A systematic review and meta-analysis of medium-chain triglycerides effects on acute satiety and food intake

Tyler Maher et al.

Summary: Research suggests that medium-chain triglycerides (MCT) may reduce energy intake more effectively than long-chain triglycerides (LCT), but the impact on subjective appetite ratings and hormone levels is less clear. Further studies are needed to understand the mechanisms by which MCT influence energy intake.

A comparison of the satiating properties of medium-chain triglycerides and conjugated linoleic acid in participants with healthy weight and overweight or obesity

Tyler Maher et al.

Summary: There is inconsistent evidence for greater satiety after consuming medium-chain triglycerides (MCT) compared to long-chain triglycerides (LCT). This study compared appetite responses in individuals with healthy weight and overweight/obesity after consuming MCT or conjugated linoleic acid (CLA). The results showed that MCT reduced energy intake over a 48-hour period, possibly due to delayed gastric emptying and increased beta-hydroxybutyrate concentrations.

Plasma GDF15 levels are similar between subjects after bariatric surgery and matched controls and are unaffected by meals

Christoffer Martinussen et al.

Summary: Plasma GDF15 does not increase in response to a liquid meal or ingestion of sucrose, isomaltose, glucose, fat, or protein. Fasting and postprandial plasma GDF15 concentrations are not increased in individuals with previous bariatric surgery compared with unoperated BMI-matched controls.

AMPK mediates energetic stress-induced liver GDF15

Logan K. Townsend et al.

Summary: The study found that AMPK plays a role in mediating the induction of GDF15 in mouse liver under conditions of energetic stress, independent of ER stress. AMPK activators can increase the expression and secretion of GDF15, suppressing food intake.

Pharmacological but not physiological GDF15 suppresses feeding and the motivation to exercise

Anders B. Klein et al.

Summary: Prolonged endurance exercise increases circulating GDF15 levels in humans to levels observed in pathophysiological conditions, but this exercise-induced GDF15 does not affect post-exercise food intake or exercise motivation like pharmacological GDF15 does.

The Role of GDF15 as a Myomitokine

Kornelia Johann et al.

Summary: Growth differentiation factor 15 (GDF15) is a cytokine that affects systemic energy metabolism and its expression and secretion in various organs and tissues are induced in different physiological and pathophysiological states, often associated with mitochondrial stress. GDF15 is considered as a myokine and cardiokine, and activation of the GDF15-GFRAL pathway may lead to increased sympathetic outflow and activation of the HPA stress axis.

GDF15: emerging biology and therapeutic applications for obesity and cardiometabolic disease

Dongdong Wang et al.

Summary: Growth differentiation factor 15 (GDF15) expression is induced by cellular stress, metformin, and exercise. Its effects on food intake and inflammation are mediated through specific receptors and signaling pathways, making it a promising target for the treatment of metabolic diseases such as obesity and diabetes.

Summary: The study found that substituting a small amount of dietary long-chain fatty acids with medium-chain fatty acids in a hypercaloric high-fat diet can protect against insulin resistance and promote glucose metabolism.
