Novel compound heterozygous mutations in UHRF1 are associated with atypical immunodeficiency, centromeric instability and facial anomalies syndrome with distinctive genome-wide DNA hypomethylation

Motoko Unoki et al.

Summary: This study identified UHRF1 as a novel causative gene of Immunodeficiency, centromeric instability and facial anomalies (ICF) syndrome. By analyzing a patient with atypical symptoms, two previously unreported mutations in the UHRF1 gene were identified, which led to abnormal methylation and protein conformation changes. Through further analysis of patient cells and protein interactions, the importance of UHRF1 in the regulation of CG methylation was confirmed.

Somatic Reversion of a Novel IL2RG Mutation Resulting in Atypical X-Linked Combined Immunodeficiency

Yujuan Hou et al.

Summary: This study reports three brothers with low-normal lymphocyte counts and susceptibility to respiratory infections and cutaneous warts. Genetic analysis revealed a novel mutation in the IL2RG gene. Functional characterization showed impaired T-cell function and somatic reversion in certain cell populations. The atypical SCID phenotype in these brothers is due to a combination of hypomorphic IL2RG function and somatic reversion.

Lessons Learned From Five Years of Newborn Screening for Severe Combined Immunodeficiency in Israel

Atar Lev et al.

Summary: This study provides a 5-year summary of the SCID newborn screening program in Israel, demonstrating its effectiveness and outcomes. The program achieved high specificity and sensitivity in detecting SCID cases, resulting in the diagnosis and management of 32 SCID infants. The study highlights the importance of implementing global SCID newborn screening programs and emphasizes the value of accumulating knowledge from screening data.

Severe Combined Immunodeficiency (SCID) Screening in Arizona: Lessons Learned from the First 2 Years

Natalie A. Booth et al.

Summary: This study retrospectively reviewed the abnormal SCID newborn screening tests in Arizona from January 1, 2018, to December 31, 2019, and found an incidence of 1 in 22,819 live births. Among the diagnosed infants, four had Artemis-type SCID. The study also identified infants with congenital syndromes associated with T cell lymphopenia, with a higher representation of infants of Hispanic ethnicity. The increased incidence of SCID in Arizona is likely due to the state's unique population profile, particularly with a higher percentage of Native American and Hispanic populations.

PIDTC centers had varied pre-HSCT management practices, but there was more consistency in the use of immunoglobulin replacement and antimicrobial prophylaxis.

Parents' Perspectives and Societal Acceptance of Implementation of Newborn Screening for SCID in the Netherlands

Maartje Blom et al.

Summary: The majority of parents expressed support for NBS for SCID from both a public health and personal perspective, with emphasis on the emotional impact of an abnormal screening result. The study highlights the importance of uniform follow-up protocols and adequate information provision to address (long-term) stress and anxiety experienced by parents during and after referral. Moving towards universal SCID screening for all infants requires a close partnership of stakeholders, immunologists, geneticists, and pediatricians-immunologists in different countries.
