Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Nicotinamide (niacin) supplement increases lipid metabolism and ROS-induced energy disruption in triple-negative breast cancer: potential for drug repositioning as an anti-tumor agent

Minsun Jung et al.

Summary: Metabolic dysregulation is a key characteristic of cancer. In this study, the researchers found that NAM can regulate the metabolism of triple-negative breast cancer cells and induce cell death through bifurcating metabolic pathways. These findings provide new insights into the repositioning of NAM as an anti-metabolic agent for TNBC treatment.

A Novel Five-Gene Signature Related to Clinical Outcome and Immune Microenvironment in Breast Cancer

Yi Yang et al.

Summary: Breast cancer is the most common cancer in women and the leading cause of cancer-related deaths worldwide. Identifying biomarkers for predicting patient survival and guiding treatment decisions is crucial in providing individualized treatment plans. This study analyzed gene expression levels and clinical data to identify differentially expressed genes associated with breast cancer. Patients in the high-risk group had significantly worse overall survival, increased activity in cancer-related pathways, and decreased levels of immune cells. Additionally, KIF4A was found to be highly expressed in breast cancer cells and its suppression inhibited cell proliferation. The five-gene signature developed in this study could serve as a prognostic factor for breast cancer and guide immunotherapy selection.

A novel lactate metabolism-related signature predicts prognosis and tumor immune microenvironment of breast cancer

Zhihao Zhang et al.

Summary: In this study, a novel prognostic signature based on three lactate metabolism-related genes (LMRGs) was constructed for breast cancer (BC) patients. The results showed that the signature was closely related to the immune microenvironment, providing new insight into future anticancer therapies. The findings suggest that this signature could be used to predict the prognosis of BC patients and guide personalized treatment strategies.

Summary: TAMs are crucial immune cells infiltrating the tumor microenvironment and play complex roles in tumor development, including promoting angiogenesis, invasion, and metastasis, as well as creating an immunosuppressive environment. Targeting TAMs has shown promising results in tumor therapy, with strategies such as inhibiting angiogenesis, blocking immune checkpoints, and enhancing macrophage phagocytosis.

NAD+ Metabolism Maintains Inducible PD-L1 Expression to Drive Tumor Immune Evasion

Hongwei Lv et al.

Summary: The study reveals that NAMPT enzyme drives PD-L1 expression in various tumors and governs tumor immune evasion through CD8(+) T cell regulation. NAD(+) metabolism maintains the activity and expression of Tet1, which plays a critical role in PD-L1 expression on tumors.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Macrophage Biology and Mechanisms of Immune Suppression in Breast Cancer

Anita K. Mehta et al.

Summary: Macrophages play a significant role in breast cancer progression, with their infiltration associated with poor prognosis. Tumor-associated macrophages impede productive tumor immunity by limiting antigen presentation and supporting tumor cell survival, angiogenesis, and metastasis, highlighting the urgent need to overcome their immune suppression for durable anti-tumor immunity in breast cancer.

Summary: This study comprehensively investigated the role of immune cell infiltration patterns in tumor progression and clinical outcomes in breast cancer. Different immune infiltration patterns were identified, with high ICI scores suggesting immune-exhausted phenotype and low ICI scores corresponding to immune-activated phenotype with potential immunotherapeutic advantage. The findings contribute to a better understanding of the complexity of the tumor immune microenvironment and could potentially optimize precision immunotherapy.
