Upregulation of SLC12A3 and SLC12A9 Mediated by the HCP5/miR-140-5p Axis Confers Aggressiveness and Unfavorable Prognosis in Uveal Melanoma

Summary: In this study, the role of solute carriers (SLCs) in uveal melanoma (UVM) was investigated using an integrative multiomics analysis. It was found that high expression of SLC12A3 and SLC12A9 was associated with unfavorable prognosis in UVM patients. The study also identified the HCP5-miR-140-5p axis as a potential noncoding RNA pathway upstream of SLC12A3 and SLC12A9, which may be involved in immunomodulation and serve as a novel predictor for clinical prognosis and immunotherapy responsiveness. These findings provide valuable insights for the development of SLC-targeted therapy and drug discovery for UVM.

Summary: This study found that the use of MAGL inhibitor AM9928 can effectively block the adhesion and migration of TNBC cells, reduce the secretion of pro-inflammatory factors by cells, and inhibit the activation of HBMECs and cytokine secretion caused by TNBC. Animal experiments showed that AM9928 can inhibit the growth of mammary tumors in the mammary fat pads and reduce the permeability changes of the blood brain barrier, thus reducing the metastasis of TNBC in the brain. These findings suggest that MAGL inhibitors have the potential to be a novel strategy for TNBC treatment.

Landscape of Infiltrated Immune Cell Characterization in Uveal Melanoma to Improve Immune Checkpoint Blockade Therapy

Summary: Our study conducted a systematic analysis of infiltrated immune cell characteristics in UM, identifying three subtypes and calculating TIC scores to characterize immune patterns. Patients with high TIC scores and low BAP1 mutations and high EIF1AX mutations showed better prognosis. High TIC score patients exhibited immune inflammatory response activation and increased expression of immune checkpoint-related genes, leading to improved prognosis and response to immunotherapy.
