Inhibition of PAD4-mediated NET formation by cl-amidine prevents diabetes development in nonobese diabetic mice

Summary: This study found that the PAD4 inhibitor cl-amidine can effectively prevent the development of T1D. The results showed that cl-amidine not only inhibits the formation of NETs in vitro and in vivo, but also delays the onset of T1D and reduces the incidence of the disease. In addition, cl-amidine reduces the production of various T1D autoantibodies, improves pancreatic inflammation, increases the frequency of regulatory T cells in pancreatic lymph nodes, and improves gut barrier dysfunction. These findings suggest that inhibition of PAD4 and blockade of NET formation may be a potential approach for the clinical treatment of T1D.

Abnormalities in lymphatic stromal components and lymph node microbiome were common in IBD and could be targeted therapeutically.

Neutrophil Extracellular Traps Caused by Gut Leakage Trigger the Autoimmune Response in Nonobese Diabetic Mice

Summary: Increased formation of neutrophil extracellular traps (NETs) is associated with gut leakage in type 1 diabetes (T1D). This study reveals that microbial alterations and loss of gut barrier function contribute to abnormal formation of NETs in NOD mice, which in turn triggers pancreatic autoimmunity. PAD4-dependent NET formation aggravates gut barrier dysfunction, activates enteric autoimmune T cells, and causes self-damage in the pancreas. These findings suggest that targeting NET degradation or inhibition may be a potential therapeutic intervention for T1D.
