Host Defense Peptides: Dual Antimicrobial and Immunomodulatory Action

Matthew Drayton et al.

Summary: The rapid rise of multidrug-resistant bacteria has raised concerns about the effectiveness of traditional antibiotics. Antimicrobial peptides, with their diverse mechanisms of actions, offer a promising solution to combat these pathogens. This review discusses the mechanisms of antibiotic resistance and the potential of antimicrobial peptides in providing a more robust treatment of bacterial infections.

Mechanism of Antimicrobial Peptides: Antimicrobial, Anti-Inflammatory and Antibiofilm Activities

Ying Luo et al.

Summary: Antimicrobial peptides (AMPs) are considered a new generation of antibiotics with not only antimicrobial activity but also antibiofilm, immune-regulatory, and other activities. Research on the mechanism of action of AMPs can aid in their modification and development, with many studies focusing on this area. AMPs exhibit a dual mechanism of action and possess antifibroblast and anti-inflammatory properties, making them a promising therapeutic prospect.

Marine Peptide-N6NH2 and Its Derivative-GUON6NH2 Have Potent Antimicrobial Activity Against Intracellular Edwardsiella tarda in vitro and in vivo

Huihui Han et al.

Summary: In this study, four derivatives of marine peptide-N6NH2 were designed and GUON6NH2 showed superior stability, lower toxicity, longer post-antibiotic effect, higher intracellular inhibition rate against Edwardsiella tarda, suggesting it as a potential novel antimicrobial agent.

Antibiotic-cell-penetrating peptide conjugates targeting challenging drug-resistant and intracellular pathogenic bacteria

Samantha M. Zeiders et al.

Summary: The failure to treat bacterial infections is a current threat due to resistance mechanisms, but using cell-penetrating peptides (CPPs) to enhance antibiotics has shown increased permeation and selectivity against both sensitive and drug-resistant pathogens. This strategy has the potential to improve antibiotic effectiveness and repurpose existing drugs for broader use against various bacterial strains.

Gran1: A Granulysin-Derived Peptide with Potent Activity against Intracellular Mycobacterium tuberculosis

Reiner Noschka et al.

Summary: Gran1, a synthetic fragment of Granulysin, demonstrates efficient antimicrobial activity against Mycobacterium tuberculosis and clinically relevant non-tuberculous mycobacteria. It interacts with the surface of Mtb, causing lethal distortions of the cell wall, while showing no off-target effects in primary human cells or zebrafish embryos. Gran1 is selectively internalized by macrophages, restricting the proliferation of the pathogen, and its hypothesis-driven design serves as a powerful approach for identifying small bioactive compounds with specific antimicrobial activity.
