Colon stroma mediates an inflammation-driven fibroblastic response controlling matrix remodeling and healing

Summary: By performing single-cell RNA sequencing on stromal cells derived from the colon, this study identified different classes of fibroblasts with distinct gene signatures regulated by chronic inflammation, including IL-11-producing inflammatory fibroblasts. The study also revealed the transcriptional program associated with trans-differentiation of mucosa-associated fibroblasts and defined a functional gene signature linked to matrix deposition and remodeling in the inflamed colon. The critical role of metalloprotease Adamdec1 at the interface between tissue remodeling and healing during colitis was demonstrated, highlighting its requirement for colon epithelial integrity.
