Prostaglandin PGE2 Receptor EP4 Regulates Microglial Phagocytosis and Increases Susceptibility to Diet-Induced Obesity

Summary: In rodents, activation of microglia through EP4 signaling promotes weight gain and insulin resistance during high-fat-diet (HFD) feeding. Microglial EP4 deletion in a specific mouse model resulted in reduced weight gain, food intake, and improved insulin sensitivity. Mechanistically, EP4-deficient microglia showed attenuated phagocytic state and decreased interaction with pro-opiomelanocortin (POMC) neurons, leading to increased density of POMC neurites extending into the paraventricular nucleus (PVN).
