TRAF3: A novel regulator of mitochondrial physiology and metabolic pathways in B lymphocytes

Summary: Mitochondria, the crucial organelle for cell survival and metabolism, play a significant role in cancers and can be targeted for therapy. Proteins controlling mitochondrial fission and fusion are essential for maintaining mitochondrial function, quality control, and cell survival. Recent research has identified the interaction between the mitochondrial fission factor MFF and the tumor suppressor TRAF3, revealing its involvement in regulating mitochondrial physiology and metabolic pathways in B lymphocytes and B cell malignancies. This finding provides insight into potential clinical implications and is discussed in this review.

Summary: In this study, the researchers found that the expression of IL-9 receptor (IL-9R) was significantly increased in mouse follicular B cells lacking TNFR-associated factor 3 (TRAF3), a critical regulator of B cell survival and function. They discovered a novel pathway in which TRAF3 suppresses B cell activation and Ig isotype switching by inhibiting IL-9R-JAK-STAT3 signaling. These findings provide new insights into understanding and treating human diseases involving abnormal B cell activation, such as autoimmune disorders.
