Understanding the squamous cell carcinoma immune microenvironment

Vahide Saeidi et al.

Summary: Primary cutaneous squamous cell carcinoma (cSCC) is the second most common human cancer with a rising incidence of about 1.8 million in the United States annually. Surgery is usually curative, but in some cases, cSCC can metastasize and result in death. The introduction of checkpoint inhibitor immunotherapy has significantly improved treatment outcomes for advanced or metastatic cSCC.

The liver cancer immune microenvironment: Therapeutic implications for hepatocellular carcinoma

Romain Donne et al.

Summary: The liver is a common site for primary cancer and a major cause of cancer death. Hepatocellular carcinoma (HCC) accounts for the majority of liver cancers. Immune-checkpoint inhibitors have shown promise in the treatment of HCC, but further understanding of the tumor microenvironment and the development of additional biomarkers are needed. Other immune-targeting strategies are also being developed. This review provides an overview of the HCC immune microenvironment and current immunotherapies.

Disease-modifying therapies for relapsing/active secondary progressive multiple sclerosis - a review of population-specific evidence from randomized clinical trials

Antonios Bayas et al.

Summary: Although the understanding of SPMS is evolving, early detection of relapse-independent progression remains difficult. The term RMS includes RRMS and SPMS with relapses, and disease-modifying therapies approved for RMS or active SPMS are used. However, there are differences in efficacy and safety evidence between drugs approved for RMS and SPMS. Clinical trials have shown that siponimod and interferon-beta 1b s.c. have significant reductions in disability progression in SPMS populations, but data on other drugs approved for RMS are limited to small subgroups and the potential to reduce relapse-independent progression is unclear.

Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response

Jingjing He et al.

Summary: Personalized immunotherapy targeting tumor-specific antigens (TSAs) can be achieved by activating T cells specific for these antigens, but precise prediction of neoantigens from tumor mutations is challenging. This study identified a specific type of T cells (Tas cells) in tumors that can be targeted for personalized immunotherapy. The level of Tas cells in tumors can accurately predict the response to immunotherapy.

Spatial Organization and Prognostic Significance of NK and NKT-like Cells via Multimarker Analysis of the Colorectal Cancer Microenvironment

Juha P. Vayrynen et al.

Summary: This study developed a multiplexed immunofluorescence assay coupled with digital image analysis and machine learning to identify different lymphocytic populations in colorectal cancer. The study found that T cells were more abundant than NK and NKT-like cells in colorectal cancer. NK cells expressed high levels of a cytotoxic marker and were located closer to tumor cells, while T and NKT-like cells expressing the marker were enriched in cells closer to tumor cells. Higher densities of T and NKT-like cells were associated with longer cancer-specific survival, as well as higher densities of GZMB(+) and FCGR3A(+) NK cells and closer proximity of T and NKT-like cells to tumor cells.

Summary: Hepatocellular carcinoma (HCC) is a significant complication of chronic liver disease with various treatment strategies including surgery, loco-regional techniques, and chemotherapy. Natural killer (NK) cells play a vital role in fighting tumors and infections, but deviations in their function have been observed in HCC patients. The rate of recurrence of tumor-infiltrating and circulating NK cells is associated with improved survival in HCC, indicating the importance of NK cell dysfunction in HCC progression.

Percentage of Natural Killer (NK) Cells in Peripheral Blood Is Associated with Prognosis in Patients with Gastric Cancer: A Retrospective Study from a Single Center

Ming-zhi Xie et al.

Summary: In this study involving 180 gastric cancer patients, it was found that patients with a higher percentage of NK cells had longer survival times and could serve as an independent prognostic indicator for gastric cancer. NK cell percentage was positively correlated with lymphocyte count and albumin, while negatively correlated with CA125 and neutrophil-lymphocyte ratio. NK cells showed predictive value for the 5-year survival rate of gastric cancer, especially when combined with neutrophil-lymphocyte ratio. Patients at early T, N, and clinical stages tended to have a higher percentage of NK cells compared to those at advanced stages.

Memory-like Differentiation Enhances NK Cell Responses to Melanoma

Nancy D. Marin et al.

Summary: The treatment of advanced melanoma is a clinical challenge, and natural killer (NK) cells show promise as a cellular therapy. Cytokine-induced memory-like (ML) differentiation enhances NK-cell responses against blood cancer targets, but its activity against solid tumors like melanoma needs further research. In preclinical studies, ML NK cells from healthy donors and melanoma patients exhibited increased IFN gamma production and cytotoxicity against melanoma targets, with a response partially dependent on activating receptors NKG2D and NKp46. Furthermore, in mouse models, ML NK cells showed better control of melanoma compared to conventional NK cells.
