Profiling and functional characterization of maternal mRNA translation during mouse maternal-to-zygotic transition

Chunxia Zhang et al.

Summary: Transcriptional regulation is important in early mouse embryo development. Using a new technique, the mRNA translation landscape of mouse preimplantation embryos was revealed, showing the critical role of active translation of maternal mRNAs in maternal-to-zygotic transition. The study also identified two maternal factors required for chromatin reprogramming and zygotic genome activation.

Ultrasensitive Ribo-seq reveals translational landscapes during mammalian oocyte-to-embryo transition and pre-implantation development

Zhuqing Xiong et al.

Summary: Researchers used low-input Ribo-seq technology in combination with PAIso-seq to investigate the translational landscape during oocyte-to-embryo transition in mammals. They found that translation activation is closely associated with polyadenylation, while translation repression is similar to global de-adenylation.

Translatome and transcriptome co-profiling reveals a role of TPRXs in human zygotic genome activation

Zhuoning Zou et al.

Summary: This study utilized Ribo-lite and mRNA sequencing to analyze the transcriptome and translatome of human oocytes and early embryos. The results showed widespread differential translation during the oocyte-to-embryo transition (OET) and zygotic genome activation (ZGA), functioning in epigenetic reprogramming, transposon defense, and small RNA biogenesis. Additionally, TPRX1/2/L were identified as critical regulators of ZGA.

Revisiting poly(A)-binding proteins: Multifaceted regulators during gametogenesis and early embryogenesis

Long-Wen Zhao et al.

Summary: Post-transcriptional regulation in gametes faces a unique challenge, with gene expression during gamete maturation and early cleavage dependent on existing mRNA coordination. Poly(A) tails and poly(A)-binding proteins (PABPs) play crucial roles in defining mRNA fate during this process, with distinct groups of PABPs lacking catalytic activity but involved in diverse roles through interactions with multifunctional partners. Current research focuses on understanding how germ cell-specific expressed PABPs potentially impact human fertility.

Regulation of the mammalian maternal-to-embryonic transition by eukaryotic translation initiation factor 4E

Yan Li et al.

Summary: The translation initiation factor eIF4E plays a crucial role in cap-dependent translation and the transition from maternal to embryonic control of mouse embryo development. Inhibiting eIF4E activity leads to developmental arrest at the two-cell stage, highlighting the importance of eIF4E in early embryonic transitions. This study also shows that mTOR activity and the negative regulator 4E-BP1 are involved in regulating eIF4E activity during embryonic development.

Global profiling of RNA-binding protein target sites by LACE-seq

Ruibao Su et al.

Summary: The LACE-seq method developed by Xue and colleagues allows for comprehensive profiling of RNA-binding protein targets at single-nucleotide resolution in low-input cells, shedding light on the essential functions of RBPs in germline and early embryo development. The precise mapping of RBP-binding sites in low-input cells opens up new avenues for studying the roles of RBPs in embryonic development and reproductive diseases.
