Collective Variables for Conformational Polymorphism in Molecular Crystals

Oren Elishav et al.

Summary: Controlling polymorphism in molecular crystals is challenging due to long time scales. We propose an efficient method for identifying collective variables that promote transitions between conformational polymorphs. Using this method in simulations, we observe transitions between all solid polymorphs of CL-20 and identify previously unknown intermediate forms.

ROY: Using the Method of Molecular Voronoi-Dirichlet Polyhedra to Examine the Fine Features of Conformational Polymorphism

Viktor N. Serezhkin et al.

Summary: Crystal chemical analysis of the polymorphs of ROY was conducted using the molecular VD polyhedra method. Each ROY molecule was found to have a unique conformation. A method for quantifying the significance of noncovalent interactions was proposed and successfully tested. The analysis revealed larger differences in noncovalent interactions between different forms of ROY than between different structural solutions of the same form.

The unexpected discovery of the ninth polymorph of tolfenamic acid

Pietro Sacchi et al.

Summary: A new metastable polymorph of Tolfenamic acid (TFA), form IX, was discovered unexpectedly during crystalization from isopropanol at low temperatures. Experimental and computational analysis confirmed that form IX has a stability close to the well-known forms I and II.

Advancing the use of Voronoi-Dirichlet polyhedra to describe interactions in organic molecular crystal structures by the example of galunisertib polymorphs

Viktor N. Serezhkin et al.

Summary: By using the method of molecular Voronoi-Dirichlet polyhedra, all types of atomic interactions in ten known polymorphs of the drug galunisertib (GAL) were quantitatively analyzed, highlighting the unique set of intra- and intermolecular noncovalent interactions in each polymorph. A new (RF, d) distribution diagram was introduced to emphasize the differences in noncovalent interactions of polymorphic modifications and to visually support the previously introduced k-Phi criterion. Hydrogen bonding motifs in GAL polymorphs were compared with nine polymorphs of the pharmaceutical aripiprazole, another prolific polymorphic system.
