Decoding the multicellular ecosystem of vena caval tumor thrombus in clear cell renal cell carcinoma by single-cell RNA sequencing

Yue Shi et al.

Summary: This study provides a comprehensive understanding of the mechanisms involved in tumor thrombus formation in clear cell renal cell carcinoma (ccRCC). The results show that tumor thrombus is marked by the presence of CD8(+) T cells with an exhausted phenotype and enhanced remodeling of the extracellular matrix. In contrast, proinflammatory states and increased expression of immunosuppressive markers are found in macrophages and malignant cells from primary tumors. The interactions between tumor and stroma reshape the extracellular matrix in tumor thrombus, which is associated with poor survival.

New Horizons in the Treatment of Osteosarcoma

Paul S. Meltzer et al.

Summary: Osteosarcoma is a common malignant tumor that primarily affects individuals aged 10 to 30, and is usually treated with surgery and chemotherapy. While whole-genome sequencing can detect genetic alterations, it has not yet resulted in improved treatment outcomes.

Summary: VTT is more common in patients with pelvic osteosarcoma compared to other primary bone sarcomas, and it is associated with specific factors such as LDH levels and sites of invasion. The presence of VTT significantly affects the survival time and risk of metastasis for patients.

Advancing therapy for osteosarcoma

Jonathan Gill et al.

Summary: Osteosarcoma, the most common primary bone cancer in children and young adults, presents challenges in treatment due to its rarity and molecular heterogeneity. Collaborative efforts in biological discoveries, technologies, and therapeutic approaches are expected to advance osteosarcoma treatment and improve patient outcomes. The increasing feasibility of molecular profiling, along with the creation of robust model systems and large tissue banks, has enhanced understanding of osteosarcoma biology.

Single-Cell Transcriptomics Reveals the Complexity of the Tumor Microenvironment of Treatment-Naive Osteosarcoma

Yun Liu et al.

Summary: This study identified different cellular subtypes and key transcription factors associated with survival prognosis in osteosarcoma (OS) through single-cell RNA sequencing. Moreover, it revealed the mechanisms by which osteoblastic OS cells regulate osteolysis and angiogenesis, impacting survival prognosis. Furthermore, the study highlighted the potential of immunotherapy targeting CD8(+) T cells and macrophages for OS treatment.

Pathways of immune exclusion in metastatic osteosarcoma are associated with inferior patient outcomes

John A. Ligon et al.

Summary: The study found that osteosarcoma pulmonary metastases (PMs) exhibit immune exclusion with an accumulation of tumor-infiltrating lymphocytes at the PM interface, which produce effector cytokines. Additionally, there were alternatively activated macrophages and polymorphonuclear myeloid-derived suppressor cells in different distributions between the center and edge of the PMs. These findings suggest potential cooperative immunosuppressive mechanisms that could be targeted for successful immunotherapy.
