Immuno-genomic characterisation of high-grade serous ovarian cancer reveals immune evasion mechanisms and identifies an immunological subtype with a favourable prognosis and improved therapeutic efficacy

Jie Sun et al.

Summary: This study characterized the immunogenomic landscape of HGSOC and identified prognostic genes associated with favorable outcomes. The results suggest that tumor immunogenicity is the most significant factor related to improved clinical outcomes and response to chemotherapy in HGSOC. Additionally, multiple genomic and transcriptomic determinants of immunogenicity were identified, including copy loss of chromosome 4q and deficiencies in the homologous recombination pathway. Furthermore, an immunological subtype characterized by increased infiltration of activated CD8 T cells and decreased Tregs was associated with favorable prognosis and improved therapeutic efficacy.

Androgen receptor activity in T cells limits checkpoint blockade efficacy

Xiangnan Guan et al.

Summary: Immune checkpoint blockade has revolutionized oncology by inducing durable anti-tumour immunity. In advanced prostate cancer patients, immunotherapy treatments have largely failed. However, inhibiting androgen receptor activity can increase the sensitivity of tumor-bearing hosts to checkpoint blockade therapy by enhancing CD8 T cell function and improving responsiveness to PD-1 targeted therapy.

Chromatin accessibility profiling by ATAC-seq

Fiorella C. Grandi et al.

Summary: The ATAC-seq method allows for the detection of unique chromatin landscapes associated with specific cell types without prior knowledge of epigenetic marks or transcription factors. The Omni-ATAC protocol described in this study is optimized for various cell and tissue types and includes detailed steps for library preparation and data analysis.

Androgen receptor-mediated CD8+ T cell stemness programs drive sex differences in antitumor immunity

Chao Yang et al.

Summary: This study reveals the immunological mechanism underlying sexual differences in non-reproductive cancers, showing that male CD8(+) T cells have impaired effector and stem cell-like properties compared to female CD8(+) T cells. Androgen receptor inhibits the activity and sternness of male tumor-infiltrating CD8(+) T cells, while castration combined with anti-PD-L1 treatment restricts tumor growth in male mice. In humans, male CD8(+) T cells have fewer stem cell-like memory cells, and AR expression correlates with CD8(+) T cell exhaustion in cancer patients. These findings provide insights into sex-based immunotherapeutic strategies for cancer treatment.

Pan-cancer landscape of T-cell exhaustion heterogeneity within the tumor microenvironment revealed a progressive roadmap of hierarchical dysfunction associated with prognosis and therapeutic efficacy

Zicheng Zhang et al.

Summary: This study conducted a comprehensive analysis of T cell exhaustion (TEX) within the tumor microenvironment (TME). The researchers identified five pan-cancer TEX subsets and revealed tissue-specific patterns and prognostic implications. They also discovered previously unknown TEX-associated cancer genes and developed a machine learning-based gene signature to model the hierarchical stages of TEX.
