Biogeographical ancestry, variable selection, and PLS-DA method: a new panel to assess ancestry in forensic samples via MPS technology

Summary: Forensic scientists are using DNA analysis to infer externally visible features and biogeographical ancestry. With advancements in sequencing technologies, new tools have been developed to simultaneously analyze millions of markers. This study created a new BGA panel using over 3000 SNPs and evaluated its accuracy in inferring ancestry. The best SNPs were selected to predict biogeographical ancestry and applied in routine forensic cases using PCR amplification.

Multivariate statistical approach and machine learning for the evaluation of biogeographical ancestry inference in the forensic field

Summary: The biogeographical ancestry (BGA) of a trace or a person/skeleton refers to the component of ethnicity, constituted of biological and cultural elements, that is biologically determined. Nowadays, many individuals are interested in exploring their genealogy, and the capability to distinguish biogeographic information about population groups and subgroups via DNA analysis plays an essential role in several fields such as in forensics. This study provides an alternative method using multivariate data analysis and machine learning strategies to evaluate BGA discriminating power of unknown samples using different commercial panels. The results show that the PLS-DA method provides more robust classifications than the XGBoost method and that commercial forensic panels may not be adequate to discriminate populations at the intra-continental level.

African mitochondrial haplogroup L7: a 100,000-year-old maternal human lineage discovered through reassessment and new sequencing

Summary: Archaeological and genomic evidence suggest that modern Homo sapiens have been roaming the planet for 300-500 thousand years. A recent study found a new mitochondria haplogroup, L7, which is estimated to be around 100 thousand years old and was previously misclassified in the literature. L7 and its sister group L5 are both low-frequency relics centered around East Africa.

Sexual Dimorphism of Cranial Morphological Traits in an Italian Sample: A Population-Specific Logistic Regression Model for Predicting Sex

Summary: Despite the popularity of sex estimation methods from crania in forensic anthropology, few studies have validated their accuracy and reliability in different populations. This study analyzes the accuracy of regression models for predicting sex from cranial morphological traits in an Italian population and proposes new models specific to the population. The results highlight the significant effects of population variability on sexual dimorphism and provide important guidance for forensic anthropologists.

A Southeast Asian origin for present-day non-African human Y chromosomes

Summary: The genomes of present-day humans outside Africa originated almost entirely from a single out-migration similar to 50,000-70,000 years ago, followed by mixture with Neanderthals contributing similar to 2% to all non-Africans. Phylogenetic analyses of Y chromosome sequences point to East/Southeast Asia as the origin of all known surviving non-African male lineages.

Origin and diffusion of human Y chromosome haplogroup J1-M267

Summary: J1-M267 is a common male Y chromosome haplogroup in West Asia that originated approximately 20,000 years ago in northwestern Iran, the Caucasus, the Armenian Highland, and northern Mesopotamia. The major branch-J1a1a1-P58-evolved during the early Holocene around 9,500 years ago in the Arabian Peninsula, the Levant, and southern Mesopotamia. The distribution of haplogroup J1-M267 in southern regions of West Asia today is likely explained by the spread of Afro-Asiatic languages, mobile pastoralism in arid zones, or both events combined.
