Tenascin-C in fibrosis in multiple organs: Translational implications

Swati Bhattacharyya et al.

Summary: Systemic sclerosis is a complex disease characterized by fibrosis in the skin and multiple internal organs. Understanding its pathogenesis and progression is a pressing scientific challenge. Recent studies have identified toll-like receptor 4 (TLR4) signaling as a key driver of fibrosis in systemic sclerosis, with TLR4 sensing danger signals and promoting fibrogenesis. Tenascin-C, an extracellular matrix glycoprotein, is highly upregulated in systemic sclerosis and plays a role in driving fibroblast activation and fibrosis progression.

The Synergistic Cooperation between TGF-beta and Hypoxia in Cancer and Fibrosis

Pramod Mallikarjuna et al.

Summary: Transforming growth factor beta (TGF-beta) regulates homeostasis and immune responses in adult animals and humans. Abnormal TGF-beta signaling promotes cancer and fibrosis, while suppressing immune responses in cancer patients. Understanding the synergistic cooperation between TGF-beta and hypoxia in development, fibrosis, and cancer can guide the development of novel anti-cancer therapeutic strategies.

Summary: This study demonstrated size-based differences in pulmonary arteries in IPF for the first time, showing their detrimental effect on lung physiology. The process of EndMT might be central to these vascular remodeling changes and could serve as a potential novel therapeutic target.

In vitro and ex vivo models in inhalation biopharmaceutical research - advances, challenges and future perspectives

Mohammed Ali Selo et al.

Summary: Oral inhalation is the preferred drug delivery method for respiratory disorders, reducing systemic side effects. The high alveolar surface area, low enzymatic activity, and rich blood supply of distal airspaces offer a promising pathway to systemic circulation. Various cell and tissue-based models have been developed to mimic pulmonary biological barriers and address biopharmaceutical questions.

Adaptation to mitochondrial stress requires CHOP-directed tuning of ISR

Sophie Kaspar et al.

Summary: This study reveals the complex interplay between three transcription factors regulating mitochondrial stress response, in which CHOP acts as a regulator to attenuate prolonged ISR and prevent unfavorable metabolic alterations. The interaction between CHOP and C/EBP beta is necessary to adjust ATF4 levels and prevent overactivation of the ATF4-regulated transcriptional program, switching ISR from an acute to a chronic state.
