Membrane-bound O-acyltransferase 7 (MBOAT7)-driven phosphatidylinositol remodeling in advanced liver disease

Summary: Advanced liver diseases are a significant cause of death worldwide, and half of these deaths are due to cirrhosis complications, while the other half are caused by viral hepatitis and hepatocellular carcinoma. However, the lack of mechanistic understanding of liver disease progression and human-relevant animal models has hindered the development of therapeutic strategies. Recent studies have found that a gene variant near MBOAT7 is associated with severe liver diseases, increasing the risk of nonalcoholic fatty liver disease, alcohol-associated liver disease, and liver fibrosis. Further studies in mice have shown that Mboat7 loss of function contributes to hepatic steatosis, inflammation, and fibrosis, establishing a causal link between this enzyme and liver health in both mice and humans. These findings offer valuable insights into liver disease progression and have implications for drug discovery in this field.
