Dichotomous effects of in vivo and in vitro ionizing radiation exposure on lymphatic function

Reetu Singh et al.

Summary: On one hand, lymphatic dysfunction leads to edema and inflammation, while on the other hand, the formation of edema and inflammation causes lymphatic dysfunction. Previous studies did not detect the apoptosis of lymphatic endothelial cells (LECs) induced by irradiation, leading to the belief that lymphatic vessels are inconsequential to ionizing radiation-induced inflammatory injury. However, this study quantified the acute responses of lymphatic vessels to irradiation and found that irradiation impairs lymphatic pumping. The study also revealed that the acute effects of irradiation on lymphatic vessels are not mediated by nitric oxide-dependent pathways.

A transcriptomic dataset evaluating the effect of radiotherapy injury on cells of skin and soft tissue

Lipi Shukla et al.

Summary: Radiation therapy can cause injury to skin and subcutaneous tissue cells, leading to various complications. By comparing the transcriptional changes in different types of cells after radiation injury through genome-wide RNA-sequencing, valuable data can be obtained. Comparing these data with publicly available data from other studies on similar or different tissue types can provide further insights into the effects of radiation injury on cells.

Phase 1 Lymfactin (R) Study: 24-month Efficacy and Safety Results of Combined Adenoviral VEGF-C and Lymph Node Transfer Treatment for Upper Extremity Lymphedema

Ida-Maria Leppapuska et al.

Summary: A study combining Lymfactin (R) with microvascular lymph node transfer surgery showed potential for reducing swelling in breast cancer-related upper limb lymphedema patients. The treatment resulted in a 46% reduction in excess arm volume after a 12-month follow-up, and also improved the transport index for some patients. These findings suggest promising results for the reduction of lymphedema.

Endothelial sprouting, proliferation, or senescence: tipping the balance from physiology to pathology

Severin Muhleder et al.

Summary: Therapeutic modulation of vascular cell proliferation and migration is crucial for controlling angiogenesis in cancer and cardiovascular disease. Increasing mitogenic stimuli may not always be beneficial for angiogenesis, as it can also lead to its arrest due to intrinsic signaling feedback loops and cell cycle checkpoints. The balance between endothelial proliferation and sprouting is tightly regulated during tissue growth, but often disrupted in disease.

Summary: This study evaluated the longitudinal impact of postmastectomy radiation therapy on persistent severe lymphedema, finding that patients who received radiation therapy were more likely to develop PSL. Risk factors for PSL included regional node irradiation, lymphedema stage, excessive volume percentage, cellulitis, among others.
