VASCULAR MECHANOTRANSDUCTION

Michael J. Davis et al.

Summary: This review provides an overview of mechanotransduction in vascular smooth muscle cells (VSMCs) and endothelial cells (ECs), discussing their sensing of mechanical stimuli and transduction of mechanical signals. It covers various mechanosensors and mechanosignaling pathways involved, as well as the mechanical regulation of transcriptome and epigenetics. The relevance of mechanotransduction to health and disease, as well as the interactions between VSMCs and ECs, are also examined. The review emphasizes the need to understand the vasculature as a whole system for a comprehensive understanding and effective management of vascular diseases.

Targeting the KCa3.1 channel suppresses diabetes-associated atherosclerosis via the STAT3/CD36 axis

Xiao-Xin Jiang et al.

Summary: Blocking the KCa3.1 channel in macrophages inhibits the development of atherosclerotic lesions in diabetic ApoE(-/-) mice fed with a high-fat diet. This inhibition is achieved by reducing the uptake of oxidized low-density lipoproteins and decreasing the expression of proinflammatory factors through the STAT3/CD36 axis. This study provides insight into a potential therapeutic target for reducing the risk of atherosclerosis in diabetic patients.

Endothelial Ion Channels and Cell-Cell Communication in the Microcirculation

William F. Jackson

Summary: Endothelial cells play a significant role in microcirculation by utilizing ion channels to communicate with neighboring cells. This communication is crucial for regulating vascular dilation and contraction, as well as controlling blood flow.

Inhibition of platelet aggregation by activation of platelet intermediate conductance Ca2+-activated potassium channels

Valentina Back et al.

Summary: This study investigated the role of platelet IKCa channels in platelet function and NO generation. The results showed that activation of IKCa channels can inhibit platelet aggregation by reducing calcium signaling and granule secretion. This finding provides a new target for the development of antiplatelet drugs that limit atherothrombosis.

Endothelial shear stress signal transduction and atherogenesis: From mechanisms to therapeutics

Lei He et al.

Summary: Atherosclerotic vascular disease and its complications are leading causes of mortality globally. Studies have shown that endothelial cells are influenced by low or disturbed shear stress at curvatures and bifurcations in the arterial tree, playing a role in the formation and development of atherosclerotic plaques. Understanding mechanosensitive signaling cascades may offer new translational strategies for therapeutic intervention of atherosclerotic vascular disease.

Summary: The study showed that endothelial-dependent vasodilation is impaired in patients with Type 2 Diabetes, but treatment with SKA-31 can improve this condition by enhancing vasodilatory responses, possibly through improved endothelial Ca2+ mobilization.

Vascular smooth muscle cells in atherosclerosis: time for a re-assessment

Mandy O. J. Grootaert et al.

Summary: Vascular smooth muscle cells play a key role in atherosclerosis by forming a protective fibrous cap and exhibiting various phenotypes that can affect plaque formation and stability. They are a larger proportion of atherosclerotic plaques than previously thought and their plasticity is regulated by various mechanisms.

Statin treatment and muscle symptoms: series of randomised, placebo controlled n-of-1 trials

Summary: The study found no overall effect of atorvastatin 20 mg on muscle symptoms compared with placebo in participants who had previously reported severe muscle symptoms when taking statins. Most people completing the trial intended to restart treatment with statins. N-of-1 trials can assess drug effects at the group level and guide individual treatment.

Benzodiazepines: Uses, Dangers, and Clinical Considerations

Summary: Benzodiazepines are widely prescribed psychoactive drugs known for their sedative effects on the central nervous system. Despite their effectiveness in treating various conditions, they have a high potential for addiction and withdrawal symptoms, requiring careful tapering of the medication. Future research is needed to explore alternative methods for treating BZD withdrawal due to the increasing number of individuals seeking treatment for BZD dependence in the United States.

K(Ca)3.1 Inhibition Decreases Size and Alters Composition of Atherosclerotic Lesions Induced by Low, Oscillatory Flow

Darla L. Tharp et al.

Summary: The increased expression of K(Ca)3.1 is associated with atherosclerotic lesion development, and inhibiting this protein can reduce lesion size and content. Treatment with TRAM-34 in mice resulted in decreased collagen, fibronectin, and smooth muscle content in atherosclerotic lesions.
