Summary: Cancer stem cells (CSCs) are a small fraction of tumor cells but play a critical role in tumor initiation, progression, therapy resistance, and recurrence. Traditional CSC-targeting drugs are ineffective against bulk tumor cells and also affect normal stem cells. Nanomaterial drug delivery systems (NDDSs) can passively accumulate in tumor tissues, reducing side effects on normal organs, and can be functionalized for targeted delivery to CSCs. NDDSs have the advantage of synchronized killing of CSCs and bulk cancer cells. This material-based approach for targeting CSCs has gained significant attention recently.

Summary: Sonodynamic therapy (SDT) is a promising cancer treatment modality for glioblastoma (GBM) due to its high tissue penetration and minimal damage to normal tissues. However, the blood-brain barrier (BBB) and hypoxic microenvironment present challenges to SDT efficiency. In this study, a biodegradable nanoplatform (CSI@Ex-A) was developed, which could effectively penetrate the BBB and target cancer cells. The nanoplatform releases hydrogen peroxide (H2O2) and oxygen (O-2) to relieve tumor hypoxia, thereby enhancing the efficacy of SDT.

Summary: Nanotechnology-based chemoimmunotherapy has shown great potential for cancer treatment, but uncontrolled transportation and synergistic responses remain challenges. In this study, a self-assembled selenopeptide nanoparticle is developed to enhance tumor chemoimmunotherapy by activating natural killer (NK) cells through the oxidative metabolite of the selenopeptide. By leveraging the advantages of size reduction and deselenization driven by reactive oxygen species, this selenopeptide can deliver therapeutics, such as doxorubicin (DOX), to solid tumors and programmatically activate NK cells. In vitro and in vivo results demonstrate the synergistic effect between DOX-induced chemotherapy and selenopeptide-induced immunotherapy, leading to improved antitumor efficacy. The selenopeptide holds promise as a stimuli-responsive immune modulator for versatile biomedical applications.

Summary: Metallic and semimetallic mesoporous frameworks have unique properties and wide applications. However, semimetallic mesoporous structures are difficult to obtain using traditional methods. In this study, monodisperse mesoporous antimony nanospheres were successfully synthesized using a selective etching route, with tunable pore sizes. The antimony nanospheres showed wide absorption in the visible to second near-infrared (NIR-II) region, with promising NIR-II photothermal performance and photoacoustic signal. These multifunctional nanospheres could be used for synergistic photothermal/chemo therapy guided by NIR-II photoacoustic imaging.
