Alzheimer's-Associated Upregulation of Mitochondria-Associated ER Membranes After Traumatic Brain Injury

Rishi R. Agrawal et al.

Summary: Traumatic brain injury (TBI) can lead to neurodegenerative diseases such as Alzheimer's disease (AD) through mechanisms that remain incompletely characterized. Similar cellular metabolic alterations and modulated cleavage of amyloid precursor protein (APP) are observed in both AD and TBI models. Increased levels of C-terminal fragment of APP (C99) in AD cell models promote the formation of transient lipid raft domains in the endoplasmic reticulum (ER) known as mitochondria-associated ER membranes (MAM domains), which significantly modulate cellular lipid homeostasis. This study provides evidence that alterations in the regulation of MAM and lipid metabolic pathways may contribute to the connection between TBI and AD.

Protein synthesis inhibition and loss of homeostatic functions in astrocytes from an Alzheimer's disease mouse model: a role for ER-mitochondria interaction

Laura Tapella et al.

Summary: The deregulation of protein synthesis and ER stress/UPR have been found in astrocytes, but their relationship and role in AD astrocytes' altered homeostatic support remain unclear. This study revealed that impaired protein synthesis in AD astrocytes is associated with increased p-eIF2 alpha and decreased GADD34 levels. The phosphorylation of eIF2 alpha and impaired protein synthesis in AD astrocytes are PERK-independent and may be caused by the altered ER-mitochondria interaction.

The AMPK-MFN2 axis regulates MAM dynamics and autophagy induced by energy stresses

Yongquan Hu et al.

Summary: Energy deprivation triggers mitochondrial fission and autophagy, while also increasing the number of mitochondrial-associated ER membranes (MAMs) through AMP-activated protein kinase (AMPK) activity. AMPK translocates to MAMs and interacts with mitofusin 2 (MFN2) during energy stress, regulating autophagy and MAM dynamics. MFN2 plays a critical role in cell survival and function under energy stress, highlighting the importance of the AMPK-MFN2 pathway in cellular stress response.

GSAP regulates lipid homeostasis and mitochondrial function associated with Alzheimer's disease

Peng Xu et al.

Summary: Research has shown that GSAP plays important roles in Alzheimer's disease and Down's syndrome by regulating multiple biological pathways including protein phosphorylation, trafficking, lipid metabolism, and mitochondrial function. GSAP physically interacts with the Fe65-APP complex to regulate APP trafficking/partitioning, is enriched in the mitochondria-associated membrane, and affects lipid homeostasis through the amyloidogenic processing of APP. Deletion of GSAP can lead to an unfavorable lipid environment for AD pathogenesis, improved mitochondrial function, and rescue of cognitive deficits in an AD mouse model.
