Sex differences in chronic pain-induced mental disorders: Mechanisms of cerebral circuitry

Elevated prelimbic cortex-to-basolateral amygdala circuit activity mediates comorbid anxiety-like behaviors associated with chronic pain

Summary: Chronic pain can lead to hyperalgesia and anxiety symptoms. This study reveals that neurons in the prelimbic cortex (PrL), which project to the basolateral amygdala (PrLBLA) and the ventrolateral periaqueductal gray (PrLl/vlPAG), exhibit increased and decreased activity, respectively, during chronic pain. Optogenetic manipulation shows that suppressing the PrL-BLA circuit reverses anxiety-like behaviors, while activating the PrL-l/vlPAG circuit reduces hyperalgesia in mice with chronic pain. Mechanistic studies suggest that elevated TNF-alpha/TNFR1 signaling in the PrL leads to increased insertion of GluA1 receptors into PrLBLA neurons and contributes to anxiety-like behaviors.
