Minisci-Type Alkylation of N-Heteroarenes by N-(Acyloxy)phthalimide Esters Mediated by a Hantzsch Ester and Blue LED Light

Summary: A synthetic method for the Hantzsch ester-mediated C2-alkylation of quinolines, isoquinolines, and pyridines using N-(acyloxy)phthalimide esters (NHPI) under blue LED light has been described. The metal-free protocol provides alkylated N-heterocyclic products with high yields and can be carried out at room temperature. The introduction of a chiral phosphoric acid enables an asymmetric version of the reaction with high enantiomeric excess values. The reaction mechanism involves the excitation of an electron-donor acceptor complex and the addition of a radical species to the N-heterocycle.

Enantioselective sulfonylation using sodium hydrogen sulfite, 4-substituted Hantzsch esters and 1-(arylethynyl)naphthalen-2-ols

Summary: This study reports a catalytic asymmetric three-component reaction for the synthesis of sulfonyl-containing axially chiral styrenes. By using a photocatalyst under visible light irradiation, 4-substituted Hantzsch esters, NaHSO3, and 1-(arylethynyl)naphthalen-2-ols were used as reactants to undergo a mild radical process, leading to high yields of axially chiral (S,E)-1-(1-(alkylsulfonyl)-2-arylvinyl)naphthalen-2-ols with excellent enantioselectivity. The reaction also demonstrated excellent regioselectivity and chemoselectivity. Sodium hydrogen sulfite was utilized as a surrogate for sulfur dioxide in this organocatalytic enantioselective radical sulfonylation under photoinduced conditions.

Enantioselective Hydroalkylation of Alkenylpyridines Enabled by Merging Photoactive Electron Donor-Acceptor Complexes with Chiral Bifunctional Organocatalysis

Summary: The potential of electron donor-acceptor (EDA) complex photochemistry in visible light-induced photocatalyst-free radical reactions has recently been recognized. However, designing catalytic asymmetric reactions driven by EDA complexes remains challenging. Existing examples are limited to single activation modes with aminocatalysts or phase-transfer catalysts. In this study, chiral bifunctional hydrogen-bonding catalysis is demonstrated to enable the asymmetric reaction of an EDA complex via dual activation modes, resulting in the formation of vicinal tertiary stereocenters at the beta,gamma-positions of pyridines with high yields and good enantio- and diastereoselectivities.

Organocatalytic discrimination of non-directing aryl and heteroaryl groups: enantioselective synthesis of bioactive indole-containing triarylmethanes

Summary: This study demonstrates excellent enantioselection between aryl and heteroaryl groups using a robust organocatalytic system. By studying the versatile 2-indole imine methide, highly enantioenriched indole-containing triarylmethanes were synthesized, and the mechanism was explored through control experiments and kinetic studies. Preliminary biological studies also indicate the potential of these compounds for anticancer and antiviral drug development.

Isothiourea-catalysed transfer hydrogenation of α,β-unsaturated para-nitrophenyl esters

Summary: A new protocol has been developed for the isothiourea-catalysed transfer hydrogenation of alpha,beta-unsaturated para-nitrophenyl esters using Hantzsch ester. Good to excellent yields were observed, especially with alpha,beta-unsaturated aryl esters bearing electron-withdrawing beta-substituents. The aryl ester products can be isolated directly in moderate to excellent yields or converted to methyl esters or benzyl amides after in situ reaction with the appropriate nucleophile. Preliminary experiments showed modest enantioinduction when a chiral isothiourea catalyst was used.
