Epidemiology of esophageal cancer in 2020 and projections to 2030 and 2040

Chun-Quan Liu et al.

Summary: Esophageal cancer is a highly prevalent and fatal malignancy, with rapidly increasing cases and deaths worldwide. This study provides an analysis of the global epidemiology of esophageal cancer in 2020 and predicts future trends. The findings are crucial for global esophageal cancer clinical therapy and cancer control planning.

DIVERSITY AND BIOLOGY OF CANCER-ASSOCIATED FIBROBLASTS

Giulia Biffi et al.

Summary: Efforts to develop anti-cancer therapies have mainly targeted the epithelial compartment, but recent studies have shown the significant influence of cancer-associated fibroblasts (CAFs) in tumor progression. CAFs not only promote cancer cell proliferation, therapy resistance, and immune exclusion, but may also restrain tumor progression in certain contexts. Research on CAFs has focused on their heterogeneity, plasticity, and functions across different cancer types and states, as well as advancements in therapeutic strategies targeting CAFs currently undergoing preclinical and clinical evaluation.

MicroRNA-99 family in cancer and immunity

Joseph Eniafe et al.

Summary: The miR-99 family is a conserved group of miRs with dual roles as oncogenic miRs and tumor suppressing miRs in different types of cancer. In addition to their roles in cancer, they also play critical roles in regulating inflammatory responses and T-cell subsets.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Exosomal and intracellular miR-320b promotes lymphatic metastasis in esophageal squamous cell carcinoma

Tong Liu et al.

Summary: Cancer-cell-released exosomal miRNAs play a significant role in tumor microenvironment communication, with miR-320b identified as closely associated with lymphangiogenesis and lymphatic metastasis in ESCC patients. Through transfer by exosomes, miR-320b activates the AKT pathway in HLECs independently of VEGF-C, affecting tube formation, migration, and EMT progression in ESCC cells. Furthermore, METTL3 positively regulates pri-miR-320b maturation through an m6A-dependent mechanism.

The Key Role of Exosomes on the Pre-metastatic Niche Formation in Tumors

Xuyang Yang et al.

Summary: Exosomes released from cells transfer bio-information to recipient cells, particularly in the context of tumor pre-metastatic niche (PMN) formation, facilitating tumor cell colonization and metastasis. These exosomes induce epigenetic changes in target cells, contributing to the formation of PMN.

NETO2 promotes esophageal cancer progression by inducing proliferation and metastasis via PI3K/AKT and ERK pathway

Jia-cheng Xu et al.

Summary: Our study revealed that high expression of NETO2 in ESCC patients was associated with tumor staging and lymph node metastasis, promoting ESCC progression through enhancing cell proliferation, suppressing apoptosis, and stimulating tumor growth. Additionally, NETO2 was found to play a crucial role in regulating migration, invasion, and epithelial-mesenchymal transition in ESCC cells. Mechanistically, NETO2 activated ERK, PI3k/AKT, and Nrf2 signaling pathways, with Nrf2 identified as a critical downstream mediator responsible for tumorigenesis. This suggests that NETO2 may serve as a potential therapeutic target or prognostic biomarker for ESCC patients.
