Summary: This study aimed to investigate the heterogeneity of late-life depression (LLD) by analyzing multimodal data including neuroanatomy, cognitive functioning, clinical symptoms, and genetics. Two dimensions were identified to delineate the heterogeneity of LLD, showing distinct differences in brain anatomy, cognitive abilities, clinical phenotype, and genetic profiles. Patients in dimension 1 had relatively preserved brain anatomy, while patients in dimension 2 exhibited widespread brain atrophy, cognitive impairment, and more severe depression symptoms.
