Summary: The American Cancer Society predicts that there will be 1,958,310 new cancer cases and 609,820 cancer deaths in the United States in 2023. While incidence trends for most cancers are favorable, prostate cancer saw a significant increase of 3% annually from 2014 to 2019. However, the overall cancer death rate continues to decline due to advances in treatment, with a 33% reduction since 1991.

Microenvironment-driven sequential ferroptosis, photodynamic therapy, and chemotherapy for targeted breast cancer therapy by a cancer-cell-membrane-coated nanoscale metal-organic framework

Wei-Lun Pan et al.

Summary: In this study, a nanomedicine based on the tumor microenvironment (TME) was designed and developed for effective cancer treatment. The nanocomposite with a cancer cell membrane (CM) coating can evade the immune clearance and accumulate at the tumor site. Activation of the nanocomposite in the TME triggers a series of reactions, promoting cancer cell death and enhancing photodynamic therapy (PDT) efficacy. This sequential synergistic therapy has demonstrated the ability to inhibit cancer cell proliferation effectively.

Targeting hypoxia-inducible factors for breast cancer therapy: A narrative review

Shuang Luo et al.

Summary: Hypoxia-inducible factors (HIFs), central regulators in the adaptation of cells to low oxygen levels, are frequently overexpressed and activated in breast cancer. They influence various processes related to cancer development, such as glycolysis, angiogenesis, and metastasis, thus affecting the prognosis of breast cancer patients. Targeting HIFs specifically could potentially enhance the efficiency of cancer therapy. Additionally, drug delivery systems (DDSs) for HIF targeting, including liposomes, polymers, and nanoparticles, are being developed.

The strategy of precise targeting and in situ oxygenating for enhanced triple-negative breast cancer chemophototherapy

Manxiang Wu et al.

Summary: This study developed a core membrane nanoplatform for enhanced treatment of triple-negative breast cancer (TNBC) using biomimetic nanotechnology and tumor microenvironment (TME) responsiveness. The nanoplatform showed precise drug delivery and improved oxygen levels in the tumor, resulting in enhanced chemotherapy/phototherapy. In addition, it exhibited good fluorescence and magnetic resonance imaging performances, enabling image-guided combination tumor therapy.

Development of MnO2hollow nanoparticles for potential drug delivery applications

Allison Greene et al.

Summary: This study reported the development of novel hollow manganese dioxide nanoparticles coated with polydopamine for potential immobilization of chemical agents. Through comparison with solid manganese dioxide nanoparticles and graphene oxide sheets, the hollow manganese dioxide nanoparticles were found to degrade faster, have a higher surface area, and better colloidal dispersion. The study also showed that the novel nanoparticles can internalize cells and inhibit cell growth through particle aggregation, providing a potential groundbreaking solution for new drug delivery systems in cancer or other applications.

Tumor Hypoxia Drives Genomic Instability

Ming Tang et al.

Summary: Hypoxia is associated with poor prognosis in cancer due to enhanced tumor malignancy and therapeutic resistance. The aggressiveness of tumors partially stems from hypoxia-induced genomic instability. Understanding how tumor hypoxia induces genomic instability is crucial for improving cancer therapeutics.
