Oxidative protein folding fidelity and redoxtasis in the endoplasmic reticulum

Lei Wang et al.

Summary: Oxidative protein folding, catalyzed by ER sulfhydryl oxidase 1 (Ero1) and protein disulfide isomerase (PDI), is essential for the function of secretory cells in eukaryotic organisms. This article summarizes oxidative protein folding in yeast, plants, and mammals, and explores the regulation of the human Ero1-PDI machinery through post-translational modifications (PTMs). The maintenance of oxidative protein folding fidelity and ER redox homeostasis relies on precise control of Ero1 oxidase activity and the collaboration among PDI family members. Additionally, the deregulation of Ero1-PDI functions contributes to human diseases and can be targeted for therapeutic interventions.

The Cellular and Subcellular Organization of the Glucosinolate-Myrosinase System against Herbivores and Pathogens

Qiaoqiao Lv et al.

Summary: Glucosinolates are important secondary metabolites in Brassicales plants, playing a critical role in chemical defense. They are chemically inactive but can be hydrolyzed by myrosinases to produce toxic compounds, forming the glucosinolate-myrosinase defense system. Research has revealed that Brassicales plants have complex biosynthetic pathways for glucosinolate production and different classes of myrosinases with distinct catalytic mechanisms and substrate specificity. Progress has been made in understanding the cellular and subcellular organization of the glucosinolate-myrosinase system, especially in the detonation of the mustard oil bomb after tissue damage caused by herbivores and pathogens.

Biogenesis and Lipase-Mediated Mobilization of Lipid Droplets in Plants

Yun Ju Choi et al.

Summary: Cytosolic lipid droplets (LDs) mainly contain neutral lipids and are considered energy reserves. TAG synthesis in plants involves the cooperation of plastids and the endoplasmic reticulum, with several proteins playing important roles in LD formation. Oleosin is critical for LD integrity and stability, while SDP1 is responsible for mobilizing TAGs in LDs.

The Endoplasmic Reticulum Role in the Plant Response to Abiotic Stress

Sofia Reyes-Impellizzeri et al.

Summary: This review focuses on the impact of alterations in components of ERQC, ERAD, and UPR on plant responses to abiotic stresses. Evidence shows a clear connection between ERAD and UPR mechanisms, but the connection of ERQC components with these processes or their possible client proteins is lacking. Proteomics approaches are suggested to uncover the identity of these proteins and their connection to ER proteostasis.

The Arabidopsis transcription factor NAI1 activates the NAI2 promoter by binding to the G-box motifs

Shayan Sarkar et al.

Summary: Brassicaceae plants, such as Arabidopsis thaliana, develop ER bodies for chemical defense against herbivores, regulated by the transcription factor NAI1. NAI1 binds to specific DNA motifs in the promoters of NAI2 and BGLU23 genes, activating their expression and influencing ER body formation and function. The study demonstrates that NAI1 has motif-dependent DNA binding activity in the nucleus, suggesting its role in gene regulation within the ER body pathway.
