Sialylation regulates neutrophil transepithelial migration, CD11b/CD18 activation, and intestinal mucosal inflammatory function

Summary: Polymorphonuclear neutrophils (PMNs) are crucial for clearing microbes and promoting tissue repair, but dysregulated PMN trafficking and tissue damage are common in inflammatory mucosal diseases. The final step of PMN migration in mucosal lined organs involves transepithelial migration (TEpM). CD11b/CD18, a beta 2-integrin, plays a role in PMN intestinal trafficking, and recent studies suggest that terminal fucose and GlcNAc glycans on CD11b/CD18 can be targeted to reduce TEpM. However, the role of sialic acid (Sia) in regulating PMN epithelial influx and inflammatory function is not well understood. This study shows that inhibiting sialidase-mediated removal of alpha 2-3-linked Sia from CD11b/CD18 inhibits PMN migration across intestinal epithelium and affects PMN inflammatory effector functions. Sialidase inhibition also reduces CD11b/CD18 activation and downstream intracellular signaling. These findings suggest that targeting sialylated glycans on CD11b/CD18 could be a novel approach to reducing tissue destruction in inflammatory mucosal diseases.

Neutrophil Extracellular Traps as an Exacerbating Factor in Bacterial Pneumonia

Summary: Neutrophils release NETs to defend against microbes, but the histones in NETs can also induce inflammation and cytotoxicity. This study found that the concentration of citrullinated histone H3, a component of NETs, was elevated in bronchoalveolar lavage fluid from patients with bacterial pneumonia, and it correlated with lung inflammation, cell death, and levels of histone H4. In a neutrophilic inflammation environment, histone H4 increased lung damage, but blocking it during pneumonia did not significantly alter lung injury measures.

A survey of mastitis pathogens including antimicrobial susceptibility in southeastern Australian dairy herds

Summary: This study described the pathogen profile in cows with clinical and subclinical mastitis in southeastern Australia and examined the antimicrobial susceptibility of isolated pathogens. The most common bacteria causing clinical mastitis were Streptococcus uberis, while Staphylococcus aureus was the most common cause of subclinical mastitis. The prevalence of antimicrobial resistance among common isolates was low.

Summary: This study established an in vitro murine mammary alveolar epithelial cell-based model and an in vivo murine mastitis model to investigate the pathogenesis of Mycoplasma bovis-induced mastitis. It was found that M. bovis activated nuclear factor kappa B (NF-kB) in the epithelial cells in a dose-dependent manner, and both live and UV-treated M. bovis strains induced inflammation in the murine mammary tissues. The developed models can be used to further study M. bovis virulence factors and host immune response to intra-mammary infections.
