Revealing the Tick Microbiome: Insights into Midgut and Salivary Gland Microbiota of Female Ixodes ricinus Ticks

Anna Wiesinger et al.

Summary: Due to climate change, there has been an increase in tick activity and population, leading to a higher prevalence of tick-borne diseases. This study analyzed the microbial communities within different tick tissues and used 16S rRNA gene sequencing to study the microbiota of Ixodes ricinus ticks. The inclusion of a mock community during bioinformatic analysis proved to be useful in identifying key bacteria within the tick.

Thermostable Keystone Bacteria Maintain the Functional Diversity of the Ixodes scapularis Microbiome Under Heat Stress

Alejandra Wu-Chuang et al.

Summary: Variations in tick microbiome due to high temperatures can influence community member hierarchy in response to environmental change. High temperatures alter microbial community structure and decrease the number of keystone taxa, which are essential for stability and functional resilience of tick microbiome under heat stress. Keystones taxa play a crucial role in retaining the majority of metabolic pathways in the functional profile of tick microbiome.

A proteomics informed by transcriptomics insight into the proteome of Ornithodoros erraticus adult tick saliva

Ricardo Perez-Sanchez et al.

Summary: Using a proteomics informed by transcriptomics (PIT) approach, the researchers successfully obtained and characterized the proteome of the saliva of Ornithodoros erraticus adult ticks. They identified novel salivary proteins and functions, and found significant differences between sexes in the saliva proteome of O. erraticus ticks. These findings improve our understanding of feeding physiology in O. erraticus ticks and will contribute to the development of vaccines for controlling O. erraticus infestations and the diseases it transmits.

Vector microbiota manipulation by host antibodies: the forgotten strategy to develop transmission-blocking vaccines

Apolline Maitre et al.

Summary: Human and animal pathogens transmitted by arthropods, such as ticks and mosquitoes, are a global concern. Control measures using acaricides are threatened by resistance in vector populations. Alternative strategies that target the vector microbiota and interfere with pathogen development are encouraged. Understanding the immune functions of host antibodies and disrupting the vector microbiota open up possibilities for novel transmission-blocking vaccines.

Anti-tick microbiota vaccines: how can this actually work?

Alejandra Wu-Chuang et al.

Summary: Ticks play a key role in transmitting pathogens to animals and humans, forming an ecological unit known as the tick holobiont. The disruption of tick-microbiota homeostasis by antibiotic treatment can impact tick fitness and pathogen interactions. Introducing anti-tick microbiota vaccines as a tool to target tick microbiome and endosymbionts shows promise in manipulating tick microbiome against ticks and transmitted pathogens for disease control.

Anti-Microbiota Vaccine Reduces Avian Malaria Infection Within Mosquito Vectors

Juste Azelyte et al.

Summary: In this study, researchers found that anti-microbiota vaccines targeting Enterobacteriaceae within mosquito midguts can modulate the mosquito microbiota and significantly disrupt the development of Plasmodium relictum in its natural vector Culex quinquefasciatus. These findings suggest that anti-microbiota vaccines can be a novel tool to control malaria transmission and potentially other vector-borne pathogens.

Bacteria related to tick-borne pathogen assemblages in Ornithodoros cf. hasei (Acari: Argasidae) and blood of the wild mammal hosts in the Orinoquia region, Colombia

Juan D. Carvajal-Agudelo et al.

Summary: Interest in research on soft ticks has grown in recent decades, providing valuable insight into their role as disease vectors. This study used metagenomics-based analyses to examine the bacterial composition of Ornithodoros cf. hasei ticks and their mammalian hosts. The findings revealed the presence of potential pathogenic bacteria in both the ticks and the host blood samples.

Rickettsia helvetica infection is associated with microbiome modulation in Ixodes ricinus collected from humans in Serbia

Apolline Maitre et al.

Summary: This study analyzed and compared the microbiota of ticks attached on humans and collected in Serbia, infected with R. helvetica or uninfected. The results showed that R. helvetica infection significantly reduced the diversity of the microbiota and the connectivity of the co-occurrence network. Bacterial taxa negatively associated with Rickettsia in R. helvetica-infected ticks were identified, suggesting competition between R. helvetica and some members of the tick microbiota. The presence of R. helvetica might have a major impact on the metabolic functions of the tick microbiome.

The bacterial community of the lone star tick (Amblyomma americanum)

L. Paulina Maldonado-Ruiz et al.

Summary: The study analyzed the bacterial community of adult A. americanum ticks collected in eastern Kansas. The majority of ticks had no culturable bacteria, and those with culturable bacteria mainly carried endosymbiotic genera such as Coxiella and Rickettsia. High throughput sequencing revealed a poor midgut bacterial community in A. americanum ticks, dominated by endosymbiotic bacteria.

Proteomics informed by transcriptomics for a qualitative and quantitative analysis of the sialoproteome of adult Ornithodoros moubata ticks

Ana Oleaga et al.

Summary: This study aimed to increase data on the composition of the saliva proteome of adult Ornithodoros moubata ticks, with a focus on females, to aid in the selection of salivary antigens for tick vaccines. Using mass spectrometry approaches, SWATH-MS proved superior in identifying proteins in female saliva and revealed significant quantitative differences between male and female saliva proteomes. This new knowledge can inform the rational selection of salivary candidates as antigen targets for tick vaccine development.

Resistance to Ticks and the Path to Anti-Tick and Transmission Blocking Vaccines

Jolieke G. van Oosterwijk et al.

Summary: The medical and veterinary public health importance of ticks and tick-borne pathogens is increasing due to the expansion of geographic ranges, increasing populations, and growing incidence of diseases. The immune-mediated control of tick infestations and pathogen transmission through vaccines has become more prominent. Research on resistance mechanisms, host immune defenses, genomics, and proteomics have contributed to the development of new anti-tick vaccine antigens. Targeting pathogen reservoirs with vaccines has the potential to disrupt disease cycles and reduce exposure in humans and animals.
