Effect of pH-sensitive nanoparticles on inhibiting oral biofilms

Summary: Dental caries is a preventable infectious disease caused by interactions between oral bacteria and dietary sugars. pH-sensitive drug delivery systems have become innovative materials for caries prevention. In this study, a pH-sensitive carrier (poly(DMAEMA-co-HEMA)) was used to synthesize a nanomaterial loaded with chlorhexidine (p(DH)@CHX), which exhibited good pH sensitivity and sustained release of CHX in an acidic environment. It showed lower cytotoxicity than free CHX and had the same antibacterial effects on biofilms. In addition, it had the potential to alter oral microbiota composition and reduce caries risk.

pH-Switchable Antimicrobial Supramolecular Hydrogels for Synergistically Eliminating Biofilm and Promoting Wound Healing

Summary: This study develops pH-switchable antibacterial hydrogels based on self-assembled amphiphilic pentapeptides, which can eradicate biofilms and promote healing in infected wounds. In vitro experiments demonstrate that the hydrogels achieve antimicrobial effects through the release of pentapeptides and antibacterial oil. In vivo experiments show that the hydrogels can accelerate wound closure, reduce inflammation, and promote angiogenesis and collagen deposition.

Summary: Dental plaque, a natural biofilm, plays a crucial role in caries and periodontal disease. The pathogenicity of S. mutans is based on the formation of extracellular polysaccharides (EPS) and its survival in acidic environments. Chemodynamic therapy (CDT) is an ideal treatment modality that utilizes the biofilm microenvironment. A novel nanocomposite, ZnO2-Cu@RB NPs, has been developed for enhanced CDT, effectively killing S. mutans in acidic biofilm and reducing demineralization of apatite. Furthermore, it inhibits acid production and EPS formation, showing promising therapeutic efficacy in a rat model.
