HIPK2 in the physiology of nervous system and its implications in neurological disorders

F. Sardina et al.

Summary: HIPK2 is an evolutionary conserved serine/threonine kinase with multifunctional roles in stress response, embryonic development, and pathological conditions. It modulates multiple signaling pathways and regulates cell fate and proliferation. HIPK2 is highly expressed in the nervous systems and is involved in neurological disorders, making it a potential therapeutic target and diagnostic or prognostic marker.

Novel elasticity measurements reveal C. elegans cuticle stiffens with age and in a long-lived mutant

Mohammad Rahimi et al.

Summary: Changes in biomechanical properties have significant effects on human health. C. elegans can be used as a model for studying the molecular genetics of mammalian tissue decline. This study found that the cuticle, not the muscle, is responsible for changes in the stretchiness of C. elegans, and collagen mutations alter the integrity and elasticity of the cuticle. Aging also stiffens the cuticle under mechanical loads beyond its healthy stretched state. Long-lived daf-2 mutants are better at preventing progressive mechanical changes with age.

Chaperone networks are shaped by cellular differentiation and identity

Khairun Nisaa et al.

Summary: Chaperone expression is regulated during development to establish tissue-specific networks. However, the molecular mechanisms behind this specificity are mostly unknown. Recent evidence suggests that differentiation transcription factors play a role in rewiring the chaperone network to meet the folding demands of the proteome, which has implications for the manifestation of protein misfolding diseases in specific tissues.

Gene regulatory network inference in long-lived C. elegans reveals modular properties that are predictive of novel aging genes

Manusnan Suriyalaksh et al.

Summary: We designed a wisdom-of-the-crowds GRN inference pipeline coupled with complex network analysis to understand the organizational principles governing gene regulation in long-lived glp-1/Notch Caenorhabdities legans. Through screening 80% of regulators, we discovered 50 new aging genes, with 86% having human orthologues. The core genes essential for longevity, including those involved in insulin-like signaling (ILS), were found, indicating the predictive functionality of the GRN structure.

The landscape of molecular chaperones across human tissues reveals a layered architecture of core and variable chaperones

Netta Shemesh et al.

Summary: The authors demonstrate that the human chaperone system consists of ubiquitous core chaperones and tissue-specific variable chaperones, and perturbation of which leads to tissue-specific phenotypes. The core chaperones are significantly more abundant across tissues and important for cell survival, forming tissue-specific functional networks. This functional organization is established in development and declines with age, highlighting the importance of understanding tissue-specific differences in protein folding capacities.
