Phase-separating RNA-binding proteins form heterogeneous distributions of clusters in subsaturated solutions

Summary: Macromolecular phase separation is an important process in the formation of membraneless biomolecular condensates in cells. In this study, the presence of heterogeneous distributions of clusters in subsaturated solutions of phase-separating RNA-binding proteins was observed, contradicting classical nucleation theory. The size of clusters increased as protein concentrations approached saturation, with some clusters containing tens to hundreds of molecules. Cluster formation in subsaturated solutions and phase separation in supersaturated solutions were found to be strongly coupled, and could be decoupled using solutes and specific mutations.
