Structure of a volume-regulated heteromeric LRRC8A/C channel

Sonja Rutz et al.

Summary: Volume-regulated anion channels (VRACs) play a role in cellular response to osmotic swelling. These membrane proteins are composed of heteromeric assemblies of LRRC8 subunits, with compositions affecting permeation properties. In this study, the structure of murine LRRC8A/C channels was characterized. The assembly showed a predominant organization with an A:C ratio of two, with LRRC8A subunits stabilizing a closed state of the channel while LRRC8C subunits enhancing the activation properties.

Structures of human pannexin-1 in nanodiscs reveal gating mediated by dynamic movement of the N terminus and phospholipids

Maki Kuzuya et al.

Summary: In this study, the cryo-EM structures of human PANX1 in lipid nanodiscs were analyzed to elucidate the gating mechanism and its regulation by the amino terminus in phospholipids. The results showed that the amino-terminal domain plays an essential role in channel opening and is associated with lipid movement.

LRRC8A homohexameric channels poorly recapitulate VRAC regulation and pharmacology

Toshiki Yamada et al.

Summary: Swelling-activated volume-regulated anion channels (VRACs) are heteromeric channels comprising LRRC8A and LRRC8 paralogs. Cryo-EM structures of LRRC8A homohexamers have limitations in mimicking VRAC function, while LRRC8A + LRRC8C heteromers exhibit strong DCPIB sensitivity that is mu dependent. The LRRC8A R103F mutation eliminates voltage dependence under low intracellular mu conditions.

Human Pannexin 1 channel: Insight in structure-function mechanism and its potential physiological roles

Eijaz Ahmed Bhat et al.

Summary: Pannexins are a large non-gap junction super family in vertebrates that play multiple roles in cellular functions through ATP release. While PANXs are known for their exclusion from gap junctions and facilitation of ATP release from cells, particularly through PANX1, recent research has focused on the Cryo-EM structure of Panx1 channel and its physiological and pathophysiological implications. This review emphasizes recent advancements in understanding the structure and function of the novel heptameric channel, particularly in the context of ATP release in health and disease.

Visualization of the mechanosensitive ion channel MscS under membrane tension

Yixiao Zhang et al.

Summary: Mechanosensitive channels, such as MscS, play a crucial role in sensing mechanical forces in cell membranes. The structure of MscS in different membrane environments is dynamic and regulated by various associated lipids, including pore lipids, gatekeeper lipids, and pocket lipids. These lipids have distinct roles in MscS mechanosensation by preventing ion conduction in the closed state, stabilizing the closed conformation, and allowing the channel to transition to different states under sustained tension.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Allosteric modulation of LRRC8 channels by targeting their cytoplasmic domains

Dawid Deneka et al.

Summary: In this study, nanobodies targeting the LRR domain of LRRC8A have been developed and characterized, providing specific modulators for LRRC8 proteins and revealing the role of their cytoplasmic domains in regulating channel activity through allosteric mechanisms. Cryo-electron microscopy and electrophysiology experiments showed that these nanobodies can alter LRRC8A channel activity by binding to distinct epitopes of the LRR domain and changing channel conformations.
