TAZ links exercise to mitochondrial biogenesis via mitochondrial transcription factor A

Jun-Ha Hwang et al.

Summary: This study reveals that TAZ is a novel stimulator for mitochondrial biogenesis and exercise-induced muscle adaptation. TAZ stimulates the translation of mitochondrial transcription factor A via the Rheb/Rhebl1-mTOR pathway. The knockout of TAZ in mice results in decreased mitochondrial biogenesis, respiratory metabolism, and exercise ability.

Inhibition of mitochondrial fission protects podocytes from albumin-induced cell damage in diabetic kidney disease

Makoto Tagaya et al.

Summary: The mechanisms underlying the progression from endothelial damage to podocyte damage in diabetic kidney disease (DKD) and the development of massive proteinuria are investigated. Dynamin-related protein 1 (Drp1)-mediated regulation of mitochondrial fission in podocytes plays a role in the pathogenesis of proteinuria in DKD. Inhibition of mitochondrial fission in podocytes may represent a new therapeutic strategy for massive proteinuria in DKD.

Mechanisms of podocyte injury and implications for diabetic nephropathy

Federica Barutta et al.

Summary: Albuminuria is a significant feature in various glomerulopathies, including diabetic nephropathy. The damage to podocytes, which play a key role in maintaining the glomerular filtration barrier, is the main cause of albuminuria. Understanding the mechanisms of podocyte injury may lead to the discovery of new treatment targets and advancements in podocyte regenerative medicine.

Influence of NAFLD and bariatric surgery on hepatic and adipose tissue mitochondrial biogenesis and respiration

Julie S. Pedersen et al.

Summary: Impaired mitochondrial function in liver tissue may contribute to the pathogenesis and disease progression of nonalcoholic fatty liver disease (NAFLD). Patients with obesity have lower mitochondrial capacity in adipose tissues but higher capacity in the liver, without overall associations to NAFLD severity, and bariatric surgery increases hepatic mitochondrial respiration and mitochondrial biogenesis.

Mfn2-mediated mitochondrial fusion alleviates doxorubicin-induced cardiotoxicity with enhancing its anticancer activity through metabolic switch

Mingge Ding et al.

Summary: This study found that inhibition of Mfn2-mediated mitochondrial fusion is associated with cardiac dysfunction and tumorigenesis. Restoring Mfn2-mediated mitochondrial fusion can enhance mitochondrial oxidative metabolism, reduce cellular injury and oxidative stress, and provide a dual therapeutic advantage in Dox-based chemotherapy by defending against Dox-induced cardiotoxicity and boosting its antitumor potency.

Calpastatin prevents Angiotensin II-mediated podocyte injury through maintenance of autophagy

Imane Bensaada et al.

Summary: The study revealed that chronic angiotensin II-induced hypertension inhibits autophagy in mouse glomeruli, leading to accelerated podocyte injury when autophagy-related gene Atg5 is deleted. However, transgenic mice expressing the endogenous calpain inhibitor calpastatin showed higher podocyte autophagy and resistance to angiotensin II-induced inhibition, demonstrating a potential protective effect against podocyte damage and albuminuria. These findings suggest that preventing calpain-mediated inhibition of autophagy may offer a new therapeutic strategy for nephropathies associated with high renin-angiotensin system activity.

Astragaloside II Ameliorated Podocyte Injury and Mitochondrial Dysfunction in Streptozotocin-Induced Diabetic Rats

Jun Su et al.

Summary: Astragaloside II (AS II), a novel saponin from Astragalus membranes, shows protective effects on podocyte injury and mitochondrial dysfunction in diabetic nephropathy (DN) by ameliorating albuminuria, renal histopathology, podocyte apoptosis, and restoring the expression of mitochondrial dynamics-related and autophagy-related proteins. Furthermore, AS II increases the expression of PINK1 and Parkin associated with mitophagy, and facilitates antioxidative stress ability via regulation of Nrf2 and Keap1 pathway, providing a potential therapeutic strategy for the treatment of DN.
