SWI/SNF complex-deficient soft tissue neoplasms: An update

Summary: The SWI/SNF chromatin remodeling complex plays a key role in regulating gene expression and cell proliferation in an ATP-dependent manner. Mutations in certain subunits of the complex are associated with various soft tissue neoplasms in human cancers. Specifically, SMARCB1 is the most frequently inactivated subunit, implicated in a range of malignant tumors.

Summary: ATRTs are aggressive childhood malignancies of the central nervous system caused by inactivating mutations in SMARCB1 or SMARCA4 genes. SMARCB1 mutated ATRTs can be divided into three molecular subgroups, while ATRT-SMARCA4 represent a distinct molecular subgroup. The distinction is supported by DNA methylation and RNA sequencing analyses.
