Aqueous-phase synthesis of upconversion metal-organic frameworks for ATP-responsive in situ imaging and targeted combinational cancer therapy

Summary: In this study, nanoscaled ATP-responsive upconversion metal-organic frameworks (UCMOFs) were synthesized for co-delivery of therapeutic protein and chemodrugs, achieving targeted combinational therapy of cervical cancer. The UCMOFs were able to release the drugs triggered by intracellular ATP, leading to enhanced therapeutic efficacy. The findings provide an effective platform for targeted combinational cancer therapy and in situ imaging.

Upconverted/downshifted NaLnF4 and metal-organic framework heterostructures boosting NIR-II imaging-guided photodynamic immunotherapy toward tumors

Summary: The study developed Er3+-doped NaLnF4@MOF core@shell nanoparticles for integrating near-infrared (NIR) driven photodynamic therapy (PDT) and NIR-II imaging. The designed NaLnF4 nanoparticles exhibited simultaneous upconversion and NIR emission under a single excitation, allowing for efficient energy transfer to the MOF shell to generate 1O2. The NIR emission could be used for tracking the location and distribution of the heterostructures via NIR-II imaging. When combined with alpha-PD-L1 immunotherapy, the nanoplatform exhibited an effective tumor inhibition rate of 95%, inhibiting both primary and distant tumors.

Target delivery of photo-triggered nanocarrier for externally activated chemo-photodynamic therapy of prostate cancer

Summary: This study established a dual-mode nanocarrier for combining chemotherapy and photodynamic therapy by embedding chemotherapeutic drugs and ROS generators onto metal-organic frameworks. By enhancing targeting efficiency with a prostate cancer-specific ligand and controlling drug release with a polymer coating, a more tightly controlled release of drug was achieved. The results show potential for an NIR-based targeted PDT system with burst release of drug and ROS to improve synergistic therapeutics.
