Cyclodextrin Metal-Organic Framework as a Broad-Spectrum Potential Delivery Vehicle for the Gasotransmitters

Li-Guo Liao et al.

Summary: The important role of gasotransmitters in physiology and pathophysiology suggests using them for biomedical treatment. However, difficulties in storage and controlled delivery hinder the development of effective gasotransmitters-based therapies. Here, a gamma-cyclodextrin metal organic framework (gamma-CD-MOF) is used as a delivery vehicle for various gasotransmitters, with the release rate being tunable by modifying the gamma-CD-MOF with different Pluronics. The biological relevance of the gasotransmitters produced by this method is demonstrated by their DNA cleavage ability and anti-inflammatory effects.

Summary: Photoelectrochemical capacitors, particularly those using porous metal-organic frameworks (MOFs), have attracted increasing attention in biomedical applications. By conducting transient photocurrent measurements using PZ-MOF electrodes, we observed cathodic current growth and decay, and the generation of anodic reverse current, indicating the presence of spike and overshoot currents. However, the introduction of excess oxygen into the electrolyte eliminated these spike and overshoot currents. These findings demonstrate the potential of PZ-MOFs as a promising material for PEC capacitors in future biomedical applications.

Homotypic Targeted Photosensitive Nanointerferer for Tumor Cell Cycle Arrest to Boost Tumor Photoimmunotherapy

Xue-Feng Bai et al.

Summary: Recent advances in tumor immunotherapy primarily focus on remodeling the immunosuppressive tumor microenvironment (TME) to enhance immune responses. However, the complexity of TME limits the effectiveness of single interventions. In this study, we developed a photosensitive nanointerferer that can trigger direct antitumor immune responses by exposing intrinsic features of tumor cells. The nanointerferer delivers a photosensitizer and small interfering RNA to inhibit cyclin-dependent kinase 4 (Cdk4) expression, leading to cell cycle arrest, antigen exposure, and increased expression of programmed cell death protein ligand 1 (PD-L1). Under laser irradiation, the nanointerferer induces photodynamic damage, releases tumor antigens, and recruits dendritic cells (DCs), promoting the activity of CD8(+) T cells in combination with anti-PD-L1 antibodies. This approach effectively inhibits tumor progression in a mouse model without significant adverse effects, providing an alternative treatment option for antitumor immunity.

Supramolecular agents for combination of photodynamic therapy and other treatments

Nahyun Kwon et al.

Summary: Photodynamic therapy (PDT) shows promise in treating various cancers, but its therapeutic benefits are limited by oxygen dependence and inadequate light permeability. Recent advancements in supramolecular approaches have led to the development of multifunctional supramolecular photosensitizer assemblies that combine PDT with other therapies to achieve synergistic effects and maximize treatment outcomes.

Cancer photo-immunotherapy: from bench to bedside

Miao Wang et al.

Summary: The combination of targeted therapy and immunotherapy is considered an ideal strategy for treating metastatic cancer, with phototherapy emerging as a promising targeted therapy that can initiate antitumor immune responses through induced immunogenic tumor cell death. The use of phototherapy in conjunction with immunoadjuvant therapy has shown to induce a systemic antitumor immune response, controlling residual tumor cells at the treatment site and distant metastases.
