High throughput single-cell genome sequencing gives insights into the generation and evolution of mosaic aneuploidy in Leishmania donovani

Gabriel H. Negreira et al.

Summary: This study for the first time revealed the extent and dynamics of whole karyotype heterogeneity in clonal populations of Leishmania. As the population expands, mosaic aneuploidy increases gradually, affecting a defined subset of chromosomes.

Selective Whole-Genome Amplification as a Tool to Enrich Specimens with Low Treponema pallidum Genomic DNA Copies for Whole-Genome Sequencing

Charles M. Thurlow et al.

Summary: This study developed a selective whole-genome amplification method to improve the yields of Treponema pallidum DNA in clinical specimens and successfully identified mutations associated with syphilis resistance. This provides an effective approach for directly sequencing low quantities of syphilis strains.

Characterizing the genomic variation and population dynamics of Plasmodium falciparum malaria parasites in and around Lake Victoria, Kenya

Ashley Osborne et al.

Summary: Characterizing the genomic variation and population dynamics of Plasmodium falciparum parasites in the Lake Victoria basin is important for malaria elimination efforts. The study uses whole-genome sequencing to assess genetic diversity in the region, finding that the parasites in the Lake Victoria basin form a distinct cluster within East African populations and have ancestry from both Central and East Africa. Drug resistance biomarkers were also identified, indicating potential challenges for treatment strategies in the region.

Localized skin inflammation during cutaneous leishmaniasis drives a chronic, systemic IFN-γ signature

Camila Farias Amorim et al.

Summary: Cutaneous leishmaniasis caused by Leishmania braziliensis exhibits both IFN-gamma-mediated signaling and cytolytic responses in cutaneous lesions. RNA-seq analysis of blood samples from CL patients revealed transcriptional signatures of both responses in the peripheral blood, suggesting systemic immune responses. The presence of ISGs and elevated IFN-gamma levels in the blood indicate a mechanism to control parasite spread and pre-activate cells for parasite killing.
