DIALS as a toolkit

Graeme Winter et al.

Summary: The DIALS software is introduced as a toolkit for processing X-ray diffraction data, emphasizing its usage as a toolkit for data processing, with support for automated systems and command-line usage, and the ability to write new tools using the API for bespoke analysis. In addition to macromolecular X-ray crystallography, DIALS can also be applied to techniques outside of this field.

Structure of trypanosome coat protein VSGsur and function in suramin resistance

Johan Zeelen et al.

Summary: Recent studies have shown that trypanosome strains expressing VSGsur have higher resistance to suramin. The tightly bound VSGsur and suramin defines a structurally divergent subgroup of coat proteins, with the co-crystal structure revealing how the drug binds asymmetrically within the VSG homodimer. Loss-of-contact mutations in VSGsur significantly reduce affinity to suramin, indicating that the resistance phenotype is dependent on the binding of suramin to VSGsur.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Dynamic, variable oligomerization and the trafficking of variant surface glycoproteins of Trypanosoma brucei

Khan Umaer et al.

Summary: Studies indicate that although M1.3 VSG appears as a monomer in vitro, it behaves as an oligomer in vivo trafficking assays. Research on other VSGs shows that oligomerization may be concentration-dependent.

The establishment of variant surface glycoprotein monoallelic expression revealed by single-cell RNA-seq of Trypanosoma brucei in the tsetse fly salivary glands

Sebastian Hutchinson et al.

Summary: In this study, researchers used single-cell RNA sequencing to investigate the development of trypanosomes in the salivary glands and found a dynamic activation program of VSG gene expression in pre-metacyclic and metacyclic cells.
