Divergent Roles of α5 and β4 Nicotinic Receptor Subunits in Food Reward and Nicotine-induced Weight Loss in Male Mice

Alberte Wollesen Breum et al.

Summary: The physiological and molecular mechanisms by which nicotine affects energy homeostasis and food-motivated behaviors are not well understood. This study demonstrates the divergent roles of alpha 5 and beta 4 subunits of nicotinic acetylcholine receptors (nAChRs) in food reward and the essential role of beta 4-containing nAChRs in the weight-lowering effects of nicotine in obese mice. The study also suggests a crosstalk between incretin biology and nAChR signaling through beta 4-containing nAChRs. Further research into cholinergic neurotransmission and targeted treatments involving beta 4-containing nAChRs is encouraged.

An Optimized Mouse Brain Atlas for Automated Mapping and Quantification of Neuronal Activity Using iDISCO plus and Light Sheet Fluorescence Microscopy

Johanna Perens et al.

Summary: In recent years, advances in whole-brain immunolabelling and LSFM have led to the development of an optimized digital mouse brain atlas for more accurate and efficient registration of neuronal activity patterns. This atlas has shown to be beneficial in mapping changes in c-Fos expression and evaluating drug effects on whole-brain activity, providing a valuable tool for neuroscience research.

Safety, tolerability, pharmacokinetics, and pharmacodynamics of concomitant administration of multiple doses of cagrilintide with semaglutide 2.4 mg for weight management: a randomised, controlled, phase 1b trial

Lone B. Enebo et al.

Summary: The study aimed to evaluate the safety, tolerability, pharmacokinetics, and pharmacodynamics of the combination of Cagrilintide and Semaglutide for weight management. Results showed that concomitant treatment with Cagrilintide and Semaglutide was well tolerated with an acceptable safety profile. Larger and longer trials are needed to fully assess the efficacy and safety of this treatment combination.

Whole-brain activation signatures of weight-lowering drugs

Henrik H. Hansen et al.

Summary: The study aimed to explore whole-brain activation signatures of six different weight-lowering drug classes. Results showed that these drugs induced similar activation responses in the brain, concurrently activating multiple neural circuits, providing potential neuroanatomical targets for future anti-obesity therapeutics.
