p62/SQSTM1 in liver diseases: the usual suspect with multifarious identities

Chong Teik Tan et al.

Summary: Sequestosome-1 (SQSTM1), also known as p62, is an autophagy receptor that plays a crucial role in the pathogenesis of liver diseases such as NASH and HCC. Abundance of p62 inclusion bodies has been recognized as a biomarker for these diseases. However, the underlying mechanisms by which p62 regulates and contributes to the development and progression of these diseases remain incompletely understood. This review critically summarizes the findings from preclinical models and explores the potential of p62 as a predictive biomarker and therapeutic target for liver diseases.

Role of Cholesterol-Associated Steatohepatitis in the Development of NASH

Christian L. Horn et al.

Summary: The increasing prevalence of nonalcoholic fatty liver disease (NAFLD) and NAFLD-related cirrhosis in the United States and globally emphasizes the importance of understanding the mechanisms causing progression of hepatic steatosis to fibrosing steatohepatitis and cirrhosis. Accumulating evidence suggests that hepatic free cholesterol (FC) overload plays a key role in necroinflammation and fibrosis, contributing to the development and progression of nonalcoholic steatohepatitis (NASH).

Differential Yet Integral Contributions of Nrf1 and Nrf2 in the Human HepG2 Cells on Antioxidant Cytoprotective Response against Tert-Butylhydroquinone as a Pro-Oxidative Stressor

Reziyamu Wufuer et al.

Summary: In the past 25 years, Nrf2 has been primarily recognized for its role in regulating antioxidant, detoxification, and cytoprotective genes; however, Nrf1 is actually more essential for cell homeostasis and organ integrity during normal growth and development. This study uses different genotypic cell lines to investigate the differential yet integral roles of Nrf1 and Nrf2 in mediating antioxidant responses to oxidative stressors.
