Summary: The evolution of the Robo family genes remains unclear, but analysis suggests a tandem duplication in the vertebrate ancestor. The Robo3 paralog is under stronger purifying selection pressure in ancestors, while four sites under positive selection on the Ig1-2 domains of Robo4 may interfere with its binding to Slits ligand.
