Dye labeling for optical imaging biases drug carriers' biodistribution and tumor uptake

Sarah Schraven et al.

Summary: Biodistribution of nanocarriers with different dye tags was compared, showing that hydrophobic dyes were preferentially taken up by tumor cells. The uptake was actively mediated but not influenced by transporters. Additionally, labeling significantly impacted the tumor accumulation and biodistribution of nanocarriers, emphasizing the need for low-interference dyes and further exploration of dye tags for unbiased results.

Recent progress in NIR-II fluorescence imaging-guided drug delivery for cancer theranostics

Shubham Roy et al.

Summary: Fluorescence imaging in the second near-infrared window (NIR-II) has many advantages, making it a popular choice for imaging-guided drug delivery (IGDD) paradigms. This review discusses the recent progress in NIR-II IGDD-mediated cancer therapeutics, including design approaches, different therapies, and updated platforms. The review also highlights the innovation directions for NIR-II IGDD platforms to improve theranostic outcomes and clinical translation. This review will contribute to the development of safe and efficient NIR-II fluorescence-mediated anticancer drug delivery.

Cytarabine nanotherapeutics with increased stability and enhanced lymphoma uptake for tailored highly effective therapy of mantle cell lymphoma

Robert Pola et al.

Summary: The study demonstrates that HPMA-based araC co-polymers show significant anti-lymphoma efficacy in chronically relapsing MCL patients, with long-term disappearance of lymphomas even in cases of patients who relapsed after high-dose araC treatments. This provides a strong preclinical rationale for the use of HPMA-based araC co-polymers in induction, salvage or palliative therapy for MCL patients.

Acid-responsive HPMA copolymer-bradykinin conjugate enhances tumor-targeted delivery of nanomedicine

Enoch Appiah et al.

Summary: This study designed and synthesized a novel HPMA-based copolymer conjugate of BK, which showed tumor-selective action and improved accumulation of nanomedicines in the tumor, resulted in enhanced intratumoral blood flow and increased tumor deposition of nanomedicines.
