Precise Prediction of Promoter Strength Based on a De Novo Synthetic Promoter Library Coupled with Machine Learning

Summary: By constructing and characterizing a mutant library of Trc promoters, a synthetic promoter library was established with a wide range of intensities. Using this library, machine learning models were built and optimized, with the XgBoost model exhibiting optimal performance in predicting the strength of artificially designed promoter sequences. This work provides a powerful platform for predictably tuning promoters to achieve optimal transcriptional strength.

Enhanced Soluble Expression of Linoleic Acid Isomerase by Coordinated Regulation of Promoter and Fusion Tag in Escherichia coli

Summary: This study successfully improved the solubility of recombinant linoleic acid isomerase in Escherichia coli by exploring different combinations of promoters and fusion tags. Among the nine recombinant strains, the one containing the T7 promoter and MBP fusion tag showed the highest solubility at 86.2%. The study also investigated the optimal catalytic conditions and inhibitory substances for MBP-PAI.

Summary: As a natural biological macromolecule, gamma-polyglutamic acid (gamma-PGA) plays a significant role in medicine, food, and cosmetic industries due to its unique properties. Current methods for increasing gamma-PGA yield are not effective due to the viscosity issue. This study successfully improved the yield of gamma-PGA by using a dynamic regulation strategy based on quorum sensing.

Strengthening the (R)-pantoate pathway to produce D-pantothenic acid based on systematic metabolic analysis

Summary: Through systematic metabolic engineering, higher yielding D-pantothenic acid producing strains were obtained based on previously developed strains. Overexpression of lpd gene, disruption of non-PTS pathway, and enhancement of PTS pathway were conducted to increase precursor accumulation. Blocking pyruvate bypass pathway, enhancing (R)-pantothenic acid pathway, introducing heterologous proteins, and blocking other synthesis pathways significantly increased the yield of D-pantothenic acid to 6.33 g/L.
