Synthesis and in vitro antiproliferative activity of certain novel pyrazolo[3,4-b]pyridines with potential p38α MAPK-inhibitory activity

Summary: The novel pyrazolo[3,4-b]pyridine compounds exhibit potent antiproliferative activity against HepG2 and HeLa cells, with compounds 14a and 14d showing the most efficacy. These compounds can also induce cell cycle disturbance and apoptosis in HepG2 cells. Furthermore, compounds 9h and 14d are identified as the most efficient antiproliferative agents in a screening of the NCI 60-cell line panel. Additionally, some compounds show good inhibitory action against the cellular pathway regulator p38 alpha kinase.

These molecules exhibit unconventional AIE properties and high first-order hyperpolarizability values, suggesting nonlinear optical characteristics. Additionally, they show excellent binding affinity towards CDK2 and have promising applications in medicinal and materials chemistry.

Compound 14 exhibited the best antiproliferative and cytotoxic activities, as well as kinase inhibitory activity. It also showed potent in vivo tumor regression and elevated catalase level.

The predicted models of anti-colon cancer and anti-hepatoma activities of substituted 4-anilino coumarin derivatives using quantitative structure-activity relationship (QSAR)

Summary: This study aimed to predict the anti-colon and anti-hepatoma cancer activity of newly designed substituted 4-anilino coumarin derivatives using quantitative structure-activity relationship (QSAR). QSAR models were generated by optimizing the molecular and electronic properties data through H-GGA DFT/BPV86 method calculation. Two validated QSAR models were obtained, which successfully predicted the anticancer activity of 17 newly designed derivatives.

Development of adamantane scaffold containing 1,3,4-thiadiazole derivatives: Design, synthesis, anti-proliferative activity and molecular docking study targeting EGFR
