The rotation of the N-terminal domain with respect to the catalytic domain plays a crucial role in presenting and positioning the tRNA anticodon-stem-loop correctly in the ADAT2 active site. A founder mutation in the ADAT3 N-terminal domain, causing intellectual disability, is shown to hinder optimal presentation of the tRNA anticodon-stem-loop to ADAT2.
