Uniform thin ice on ultraflat graphene for high-resolution cryo-EM

Summary: Cryo-electron microscopy (cryo-EM) is used to visualize the atomic structure of macromolecules in their native state by embedding them in thin ice. However, controlling the uniformity of ice thickness is a challenge in high-resolution cryo-EM. In this study, we found that using ultraflat graphene (UFG) as the supporting film improves control over vitreous ice thickness. We successfully determined the 3D structures of hemoglobin, alpha-fetoprotein, and streptavidin using this method.
