Computational investigation of potent inhibitors against YsxC: structure-based pharmacophore modeling, molecular docking, molecular dynamics, and binding free energy

Summary: In Staphylococcus aureus, the GTP-binding protein YsxC plays a crucial role in protein synthesis. By using pharmacophore modeling and molecular docking, several molecules with higher binding affinities to YsxC than GTP were identified. These molecules share similar pharmacophore features of GTP and form stable complexes with YsxC through hydrogen bonds and hydrophobic interactions. These findings provide potential candidates for designing novel antimicrobial agents against S. aureus.

Biochemical characterization and structure-based in silico screening of potent inhibitor molecules against the 1 cys peroxiredoxin of bacterioferritin comigratory protein family from Candidatus Liberibacter asiaticus

Summary: In this study, the Bacterioferritin comigratory protein family 1 Cys peroxiredoxin from Candidatus Liberibacter asiaticus (CLaBCP) was biochemically studied and screened for potential antibacterial molecules. The results showed that CLaBCP has enzymatic activity and undergoes structural changes at different pH and temperature ranges. A structure-based drug design method was used to identify more efficient molecules, and molecular dynamics studies confirmed their stability.

Screening of marine natural products for potential inhibitors targeting biotin biosynthesis pathway in Mycobacterium tuberculosis

Summary: Tuberculosis remains a major global public health concern. The development of new drugs is necessary due to the emergence of drug-resistant strains of Mycobacterium tuberculosis. In this study, comprehensive computational methods were used to identify compounds that bind to essential enzymes involved in biotin synthesis and ligation of M. tuberculosis. Two compounds were found to consistently bind to the enzymes with high affinity. These compounds could potentially be used in the design of effective anti-tuberculosis therapeutics by inhibiting bacterial growth through blocking biotin biosynthesis.

Promising antibacterials for LLM of Staphylococcus aureus using virtual screening, molecular docking, dynamics, and MMPBSA

Summary: In this study, virtual screening was used to identify potent antibacterial molecules that can target the LLM protein in S. aureus. Molecular docking and dynamics simulations showed that five identified molecules had higher affinities and stable interactions with the LLM protein compared to tunicamycin. These molecules can potentially be used as inhibitors of LLM in S. aureus, and even showed higher potency than previously reported molecules.

The WHO Global Tuberculosis 2021 Report - not so good news and turning the tide back to End TB

Summary: The 2021 WHO global TB report presents a grim situation in the global epidemic of TB, including a stagnation in TB incidence decline, a decline in TB notifications, and an increase in estimated TB deaths. None of the targets set at the 2018 United Nations High Level Meeting on TB were achieved. The sub-optimal global performance in achieving TB control targets in 2020 is attributed to the COVID-19 pandemic, but TB programs were already off track before the pandemic, highlighting the fragility of the global TB response. Ending the global TB epidemic requires bold leadership, optimization of interventions, widespread coverage, addressing social determinants, and mobilization of adequate funding.

Structural-based virtual screening and identification of novel potent antimicrobial compounds against YsxC of Staphylococcus aureus

Summary: In this study, a 3-dimensional model of Ribosome biogenesis GTP-binding (YsxC) was built, and potent molecules against Staphylococcus aureus were identified. Molecular docking analysis revealed that the screened molecules had higher binding affinity than GTP. Molecular dynamics analysis confirmed the stability of YsxC-inhibitor(s) complexes. The findings provide valuable insights for designing novel compounds against S. aureus.

In-silico functional and structural annotation of hypothetical protein from Klebsiella pneumonia: A potential drug target

Drug-Repurposing Approach To Combat Staphylococcus aureus: Biomolecular and Binding Interaction Study

Structure-Based Identification of Potential Drugs Against FmtA of Staphylococcus aureus: Virtual Screening, Molecular Dynamics, MM-GBSA, and QM/MM

Summary: Staphylococcus aureus is resistant to beta-lactam antibiotics due to the presence of FmtA, an esterase that plays a key role in methicillin resistance. Computational drug screening identified ofloxacin, roflumilast, and furazolidone as potential molecules with high binding affinities to FmtA. Molecular dynamics simulations and analysis revealed that the FmtA-drug complexes are stable and the residues in the active site of FmtA are essential for forming stable protein-ligand complexes with lower energy. Further testing and modification of these screened molecules are needed to develop antimicrobial compounds against S. aureus.
