The enigmatic roles of epithelial gasdermin B: Recent discoveries and controversies

Andrei I. Ivanov et al.

Summary: Gasdermin B (GSDMB) is a unique member of the gasdermin family, exhibiting distinctive features such as lack of autoinhibition, binding preference to phosphatidylinositol phosphates and sulfatides, and the ability to promote lytic and nonlytic cellular functions. Its lack of a mouse ortholog poses challenges in in vivo mechanistic studies. GSDMB is highly expressed in epithelial cells of organs in direct contact with the external environment, playing a role in enteric infections, inflammatory bowel disease (IBD), and colorectal cancer. This review explores the peculiarities of GSDMB among gasdermin family members and the controversies surrounding its involvement in mammalian inflammatory cell death, pyroptosis, with recent findings highlighting its dual lytic and nonlytic functions in epithelial-derived contexts during gut health and disease.

Gasdermin D-mediated release of IL-33 from senescent hepatic stellate cells promotes obesity-associated hepatocellular carcinoma

Ryota Yamagishi et al.

Summary: In this study, the release mechanism of senescence-associated secretory phenotype (SASP) factors from senescent hepatic stellate cells (HSCs) was identified in a mouse model of obesity-induced hepatocellular carcinoma (HCC). IL-33 release from HSCs promoted HCC development via the activation of ST2-positive T-reg cells in the liver tumor microenvironment. These findings highlight the therapeutic potential of inhibitors of gasdermin D-mediated pore formation for HCC treatment.

Gasdermin D inhibition confers antineutrophil-mediated cardioprotection in acute myocardial infarction

Summary: The study identified GSDMD in activated leukocytes in the infarcted heart, showing that loss of GSDMD reduces infarct size, improves cardiac function, and increases survival after acute myocardial infarction. Excessive bone marrow-derived and GSDMD-dependent early neutrophil production and mobilization contribute to detrimental immunopathology after AMI. Pharmacological inhibition of GSDMD provides cardioprotection by reducing scar size and enhancing heart function post-AMI.

Hepatocyte pyroptosis and release of inflammasome particles induce stellate cell activation and liver fibrosis

Susanne Gaul et al.

Summary: The study found that hepatocyte pyroptosis, caused by NLRP3 inflammasome activation, leads to the release of inflammasome proteins and exacerbates fibrogenesis. Inhibition of caspase-1 and gasdermin D activation can prevent pyroptosis. The released inflammasomes are taken up by hepatic stellate cells, promoting inflammation and fibrosis. These findings suggest a novel mechanism for liver injury and potential therapeutic targets for chronic liver diseases.

Research indicates that gasdermin D plays a crucial role in connecting an elevated immune response to a sustained inflammation, linking it to the progression of non-alcoholic fatty liver disease to non-alcoholic steatohepatitis. Both basic and clinical studies show evidence supporting gasdermin D as a potential therapeutic target for diseases characterized by perpetuated inflammatory processes.
