Nutrition Strategies Promoting Healthy Aging: From Improvement of Cardiovascular and Brain Health to Prevention of Age-Associated Diseases

Monika Fekete et al.

Summary: An increasing number of studies suggest that diet plays an important role in regulating aging processes and modulates the development of age-related diseases. Certain diets with anti-aging action can slow down or prevent the development of cardiovascular and cerebrovascular diseases, neurodegenerative diseases, cognitive impairment, and dementia. The Mediterranean diet, incorporating fasting elements, may play a significant role in healthy aging.

Oxidative Stress in Age-Related Neurodegenerative Diseases: An Overview of Recent Tools and Findings

Dimitris Korovesis et al.

Summary: Reactive oxygen species (ROS) play a dual role in physiological conditions, acting as signaling molecules but also causing oxidative stress when accumulated excessively. Ageing and neurodegenerative diseases are associated with oxidative stress and the accumulation of damaged proteins. This review discusses the role of oxidative stress in physiology and pathology, as well as state-of-the-art techniques for detecting and quantifying ROS and oxidized proteins in live cells and in vivo. It also explores the recent research findings on the involvement of oxidative stress in neurological disorders.

Antioxidant Activity of Polysaccharides from the Edible Mushroom Pleurotus eryngii

Tania Petraglia et al.

Summary: This study evaluated the antioxidant and neuroprotective activity of an enriched polysaccharide fraction (EPF) obtained from the fruiting body of cultivated P. eryngii. The EPF showed high content of (1-3; 1-6)-beta-D-glucans and exhibited significant scavenging activities against DPPH, superoxide, hydroxyl, and nitric oxide radicals. It also demonstrated biocompatibility and reduced oxidative stress in DI-TNC1 cells. These findings suggest that polysaccharides extracted from P. eryngii could be used as functional food to enhance antioxidant defenses.

Edible Mushrooms as Source of Fibrin(ogen)olytic Enzymes: Comparison between Four Cultivated Species

Tania Petraglia et al.

Summary: This study compared the fibrin(ogen)olytic activity of four cultivated edible mushrooms and found that Pleurotus ostreatus had the highest activity. The enzymes showed maximum activity at pH 6-7 and 30-40 degrees C, and were inhibited by specific inhibitors. A new index called the Specific Fibrin(ogen)olytic Index (SFI) was proposed to specify the proportion of fibrin(ogen)olytic activity in the total proteolytic capacity. These findings suggest that mushroom extracts can be used as functional ingredients for the development of new thrombolytic agents.

Antioxidant and Anti-Inflammatory Activity of Five Medicinal Mushrooms of the Genus Pleurotus

Jan Stastny et al.

Summary: Edible medicinal mushrooms within the higher fungi group have a long history of being used for food and folk medicine. The Pleurotus genus, represented by Pleurotus ostreatus, has been widely cultivated and studied. In this study, lesser-known Pleurotus species were examined for their antioxidant and anti-inflammatory activities. Significant differences were found among the extracts, with P. flabellatus showing strong radical scavenging activity and P. ostreatus Florida demonstrating potent NF-kappa B inhibition. These findings suggest the therapeutic potential of these mushrooms for further investigation and clinical trials.

Summary: A novel polysaccharide PSP2-1 was isolated from Pleurotus sajor-caju. PSP2-1 showed neuroprotective effects on oxidatively damaged neuronal cells by improving cell viability and inhibiting apoptosis and release of cytochrome c. Additionally, PSP2-1 exerted its effects through inhibiting the phosphorylation of MAPK pathway. Animal experiments demonstrated that PSP2-1 improved oxidative stress injury and cognition in mice with aging.
